生物钟基因BMAL1及缺氧诱导因子HIF-1α影响鼻咽癌细胞的增殖、迁移 被引量:1

摘要 目的:从蛋白水平探讨生物钟基因BMAL1与缺氧诱导因子-1α(hypoxia inducible factor-1α,HIF-1α)间的关系及两者对鼻咽癌细胞增殖、迁移的影响。方法:通过感染慢病毒方式构建细胞株,蛋白质印迹分析验证感染效率及鼻咽癌细胞中BMAL1与HIF-1α间的关系;采用CCK-8细胞增殖实验检测鼻咽癌细胞增殖能力,划痕实验检测鼻咽癌细胞迁移能力。结果:与对照组相比,过表达BMAL1可以抑制鼻咽癌细胞中HIF-1α蛋白表达,沉默BMAL1可以促进鼻咽癌细胞中HIF-1α蛋白表达,BMAL1的作用机制可能与HIF-1α有关。CCK-8细胞增殖实验及划痕实验表明,过表达BMAL1或沉默HIF-1α能减弱鼻咽癌细胞增殖能力及迁移能力(P<0.05)。结论:蛋白水平BMAL1可能影响HIF-1α蛋白的表达,过表达BMAL1或沉默HIF-1α可以抑制鼻咽癌细胞增殖、迁移。 Objective:To investigate the relationship between circadian clock gene BMAL1 and hypoxia inducible factor HIF-1αat the protein level and their effects on the proliferation and migration of nasopharyngeal carcinoma cells.Methods:The cell line was constructed by infecting lentivirus.Western blot analysis was used to verify the infection efficiency and the relationship of BMAL1 gene and HIF-1αin nasopharyngeal carcinoma cells.The proliferation ability of nasopharyngeal carcinoma cells was detected by CCK-8 cell proliferation test,and the migration ability of nasopharyngeal carcinoma cells was detected by scratch test.Results:Compared with the control group,overexpression of BMAL1 can inhibit the protein expression of HIF-1αin nasopharyngeal carcinoma cells,silencing BMAL1 can promote the protein expression of HIF-1αin nasopharyngeal carcinoma cells.The mechanism of BMAL1 may be related to HIF-1α.CCK-8 cell proliferation test and scratch test showed that BMAL1 overexpressed or HIF-1αgene silencing could reduce the proliferation and migration of nasopharyngeal carcinoma cells(P<0.05).Conclusion:BMAL1 may affect the protein expression of HIF-1αin protein level.High expression of BMAL1 or silencing of HIF-1αcan inhibit the proliferation and migration of nasopharyngeal carcinoma cells.
