花菜类SSR分子标记筛选及其在品种鉴定上的应用 被引量:3

摘要 青花菜(B.oleracea var.italica)和花椰菜(B.oleracea var.botrytis)均为以花球为产品的甘蓝变种,亲缘关系较近,生产上常以花菜类来统称。为筛选出一套花菜类品种简单重复序列的鉴定引物,分三批从已公布的SSR引物中挑选出70对引物,以8份品种为材料,根据扩增多态性好、稳定性高、染色体上分布均匀等原则筛选出36对引物,用于花菜类品种鉴定。经173份品种验证,该引物组合可以很好地区分花椰菜与青花菜,能鉴定出绝大部分品种,只有3个品种相似系数达1,无条带差异,无法区分开。形态性状观测发现这3个品种差异极小,验证了本研究建立的SSR标记鉴定品种方法的准确性。36对引物中只需要10对核心引物就可以达到最大区分能力,不同组合理论分辨力不同,最大可达2.92×10^(12),推荐染色体上分布均匀的组合作为花菜类品种鉴定的最优组合,其理论分辨率为1.09×10^(12)。本研究表明SSR分子标记可以用于花菜类品种鉴定,可为今后花菜类产业化中种苗纯度和真实性鉴定及种质资源管理提供技术支撑。 Broccoli (B.oleracea var.italica) and cauliflower (B.oleracea var.botrytis) are cultivar of species Brassica oleracea,their main edible part is the flower bud.They are closely related and often as cauliflower class in production.In order to screen a set of simple repeat sequences primers for identification of cauliflower and broccoli varieties,70 primer pairs were selected from published database in three batches,and 8 varieties were used as materials.Screening was based on the principles of good amplification polymorphism,high stability,and uniform distribution on chromosome.36 primer pairs were selected for identification of cauliflower and broccoli varieties.The verification test by 173 varieties showed that these primers can distinguish cauliflower and broccoli,and can identify majority varieties.Only 3 varieties have a similarity coefficient of 1,which can’t identified by them.Among 36 pairs of SSR primers,only 10 pairs of core primers are needed to reach the maximum discriminating ability,and the maximum theoretical discrimination is 2.92×10^(12).The combination with uniform distribution on chromosomes is recommended for identification of cauliflower and broccoli varieties,and its theoretical discrimination is 1.09×10^(12).This study shows that SSR molecular markers can be used for the identification of cauliflower and broccoli varieties.This method will provide technical support for the identification of seedling purity and authenticity,and germplasm resource management of cauliflower and broccoli.
