摘要
目的观察头穴点按干预对脑缺血损伤大鼠大脑皮层TLR4/NF-κB通路的影响,探讨其减轻脑缺血损伤大鼠的炎症反应机制。方法将30只SD大鼠按随机数字表法分为假手术组、模型组、头穴点按组,每组各10只。除假手术组外,其余各组大鼠采用线栓法建立局灶性脑缺血模型。术后第1天对头穴点按组进行干预,由专业医师使用鍉针给予百会、神庭穴刺激,1d/次,共14d。末次治疗后,采用Zea-longa评分评价大鼠的神经功能缺损状况;TTC染色检测脑梗死率;ELISA检测大脑皮层TNF-α的表达;Western blot检测大脑皮层TLR4、NF-κB的表达。结果与假手术组比较,模型组的神经功能缺损评分,脑梗死率,大脑皮层中TLR4、NF-κB p65、TNF-α蛋白的表达均显著升高(P<0.05);与模型组比较,头穴点按组的神经功能缺损评分,脑梗死率,大脑皮层中TLR4、NF-κB p65、TNF-α蛋白的表达均降低(P<0.05)。结论头穴点按可促进脑缺血大鼠神经功能的恢复,具有一定的抗炎作用,其机制可能与抑制TLR4/NF-κB通路有关。
Objective To observe the effect of head acupoint pressing on TLR4/NF-κB pathway in cerebral cortex of rats with cerebral ischemia injury,and to explore the mechanism of head acupoint pressing on reducing inflammation in rats with cerebral ischemia injury.Methods Thirty SD rats were divided into sham operation group,model group and head acupoint pressing group.According to random number table,with 10 rats in each group.Except sham operation group,the other groups were established by occlusion of the middle cerebral artery(MCAO).On the 1 st day after operation,the head acupoint pressing group was applied to“Baihui”and“Shengting”,1 d/time,a total of 14 days.After the last treatment,Zea-Longa score was used to evaluate the neurological deficit of rats.TTC staining was used to detect cerebral infarction rate.The expression of TNF-αin cerebral cortex was detected by ELISA.The expression of TLR4 and nuclear NF-κB in cerebral cortex was detected by Western blot.Results Compared with the sham operation group,the neurological deficit score of the model group was significantly increased(P<0.05),the cerebral infarction rate was increased,and the protein expressions of TLR4,NF-κB p65 and TNF-αin cerebral cortex were increased(P<0.05).Compared with the model group,the neurological deficit score of the head acupoint pressing group was decreased(P<0.05),the cerebral infarction rate was significantly decreased,and the protein expressions of TLR4,NF-κBp65 and TNF-αin the cerebral cortex were decreased(P<0.05).Conclusion Head acupoint pressing can promote the recovery of neurological function in rats with cerebral ischemia,and has certain anti-inflammatory effects.The mechanism may be related to the inhibition of TLR4/NF-κB pathway.
