摘要
利用Illumina HiSeq高通量测序技术对仁扇舟蛾(Clostera restitura)5龄幼虫前、中、后肠细菌的16S rDNA V4区序列进行测序,分析不同肠段细菌群落结构组成,比较细菌群落结构的差异性。结果表明:仁扇舟蛾5龄幼虫前、中、后肠细菌共归属于26个门,78个纲,142个目,267个科,392个属。其中,优势菌门为变形菌门(Proteobacteria)、厚壁菌门(Firmicutes)、拟杆菌门(Bacteroidetes)、放线菌门(Actinobacteria)。在属水平上,主要有肠杆菌属(Enterobacter)、盐单胞菌属(Halomonas)、毛螺旋菌属(Lachnospira)、希瓦氏菌属(Shewanella)、拟杆菌属(Bacteroides)、不动杆菌属(Acinetobacter)、芽孢杆菌属(Bacillus)、节杆菌属(Arthrobacter)、罗斯氏菌属(Roseburia)、颤螺旋菌属(Oscillospira)、假单胞菌属(Pseudomonas)。从不同肠段细菌群落α多样性可知,中肠的物种多样性最高,后肠的物种丰富度最高,前肠的物种多样性、丰富度均最低。样本层级聚类及PCoA分析可知,中肠和后肠的群落结构相似性较高。LEfSe差异分析结果显示,中肠的差异细菌种类最多。仁扇舟蛾5龄幼虫前、中、后肠的细菌群落结构物种组成及丰度不同,且多样性存在差异,肠道细菌在提高宿主对环境的适应性上发挥作用。
The V4 region of 16S rDNA of the bacterial communities in the foregut,midgut and hindgut of the 5 th instar larvae of Clostera restitura was sequenced by Illumina HiSeq techniques.The bacterial community structure in different intestinal segments was analyzed,and the differences of bacterial community structure were compared.The foregut,midgut and hindgut gut bacteria of the 5th instar larvae of C.restitura were annotated into 26 phylum,78 classes,142 orders,267 families and 392 genera.Among them,the dominant phyla are Proteobacteria,Firmicutes,Bacteroidetes and Actinobacteria.At the genus level,the main genus of bacteria are Enterobacter,Halomonas,Lachnospira,Shewanella,Bacteroides,Acinetobacter,Bacillus,Arthrobacter,Roseburia,Oscillospira and Pseudomonas.According to the intestinal bacterial alpha diversity,the species diversity of the midgut was the highest,the richness of hindgut wasthe highest,and the foregut had the lowest species diversity and richness.From the analysis of sample level clustering and PCoA,the community structure similarity of midgut and hindgut was higher.LEfSe difference analysis results showed that the most discrepancebacteria species were found in the midgut.The species composition and abundance of gut bacteria in the foregut,midgut and hindgut of the 5th instar larvaeof C.restitura were different,and the species diversity was different.The intestinal bacteria of C.restitura might play a role in improving the host’s adaptability to the environment.
