过表达环状RNA circTCF25经miR-128靶向VEGF-C对膀胱癌细胞生物学行为的影响 被引量:6

摘要 目的本文旨在分析过表达环状RNA circTCF25对膀胱癌细胞生物学行为的影响,明确环状RNA circTCF25是否与膀胱癌相关。方法取于佳木斯大学附属第一医院病理科保存的未经放化疗治疗且经临床确诊的20例膀胱癌患者膀胱癌组织及癌旁组织,并培养膀胱癌细胞株T24。分析miR-128、VEGF-C在膀胱癌组织及癌旁组织中的表达情况,膀胱癌细胞做环状RNA circTCF25过表达和空载转染。分析过表达环状RNA circTCF25对miR-128、VEGF-C表达、膀胱癌细胞生长、增殖活性、克隆形成能力、迁移、侵袭的影响。结果膀胱癌组织中miR-128表达量低于癌旁组织,VEGF-C表达量、阳性表达率高于癌旁组织(P<0.05)。过表达组环状RNA circTCF25表达量高于空载组(P<0.05)。过表达组miR-128表达量低于空载组,VEGF-C表达量高于空载组(P<0.05)。双荧光素酶报告基因显示,环状RNA circTCF25靶向结合miR-128,miR-128靶向结合VEGF-C。过表达组细胞生长数、增殖活性、克隆形成能力均高于空载组(P<0.05)。过表达组细胞迁移、侵袭能力均高于空载组(P<0.05)。结论过表达环状RNA circTCF25可促进膀胱癌细胞生长、增殖活性、克隆形成能力、迁移、侵袭,其可能经miR-128靶向VEGF-C发挥上述作用。 Objective The purpose of this study was to analyze the effect of overexpression of circtcf25on the biological behavior of bladder cancer cells,and to determine whether circtcf25is related to bladder cancer.Methods Bladder cancer tissues and adjacent tissues of 20patients with bladder cancer treated without radiotherapy and chemotherapy and diagnosed clinically were cultured T24.bladder cancer cell lines The expression of miR-128、VEGF-C in bladder cancer tissues and adjacent tissues was analyzed.Cyclic RNA circTCF25overexpression and no-load transfection were performed in bladder cancer cells.The effects of overexpression ring RNA circTCF25on miR-128、VEGF-C expression,bladder cancer cell growth,proliferation activity,clonal formation ability,migration and invasion were analyzed.Results The expression of miR-128in bladder cancer tissue was lower than that in paracancerous tissue,and the VEGF-C expression and positive expression rate were higher than that in paracancerous tissue(P<0.05).The expression of annular RNA circTCF25in overexpression group was higher than that in no-load group(P<0.05).The miR-128expression in the overexpression group was lower than that in the no-load group,and the VEGF-C expression was higher than that in the no-load group(P<0.05).Double luci fer ase reporting gene shows that ring RNA circTCF25targeted binding miR-128,miR-128targeted binding VEGF-C.cell growth number,proliferation activity and clonal formation ability in the overexpression group were higher than those in the no-load group(P<0.05).cell migration and invasion ability in the overexpression group were higher than those in the no-load group(P<0.05).Conclusion Overexpression of cyclic RNA circTCF25can promote bladder cancer cell growth,proliferative activity,clonal formation ability,migration,invasion,which may play the above role through miR-128targeting VEGF-C.
