人参粉长期喂养去卵巢大鼠对其肝铁代谢影响的初探 被引量:1

Effect of Ginseng Powder on Liver Iron Metabolism in Ovariectomized Rats after Long-term Feeding

摘要 目的研究长期给予去卵巢大鼠一定剂量人参粉对其血清及肝脏铁代谢相关指标的影响。方法分别以0.5、1、3.0g/kg BW d人参粉掺入基础饲料喂饲去卵巢大鼠90d,另设只给予基础饲料的假手术及去卵巢大鼠对照组;试验结束测定血清雌激素与血清、肝脏铁代谢相关指标;对子宫进行大体及组织病理学检查。结果与假手术对照组比较,去卵巢对照组雌激素水平(201.93±19.17vs 148.51±29.75)明显降低(P<0.01)、子宫出现萎缩且脏/体比(9.628±2.695vs 1.960±0.786)明显减少(P<0.01);而给予不同剂量人参粉的去卵巢大鼠血清雌激素随人参剂量增加而升高(144.49±35.35vs 167.81±20.59vs 183.18±21.49)且子宫的萎缩程度逐渐减轻(1.952±0.527vs 3.120±1.120vs 4.379±1.602),且高剂量人参粉组与去卵巢对照组比较差异具有统计学意义(P<0.05)。各组血清铁代谢指标无明显变化;去卵巢后肝脏铁调素(Hepcidin)mRNA表达水平明显降低(11.11±2.34vs 8.66±2.14),FPN mRNA的表达水平升高(2.78±0.49vs 4.27±0.74),BMP6mRNA表达水平有所降低(4.56±0.44vs 3.93±0.73)且分别与假手术组比较差异明显(P<0.05);高剂量组人参粉Hepcidin的缓解性升高(10.82±2.40,P<0.05)及高、中剂量组的FPN mRNA缓解性降低分别与去卵巢对照组比较具有统计学意义(P<0.05);各剂量组人参粉对BMP6mRNA表达水平均未见明显影响。结论一定剂量人参粉长期喂养去卵巢大鼠能够通过调节其肝脏Hepcidin、FPN mRNA表达水平及血清雌激素水平,部分缓解雌激素下降引起的不良影响,提示了人参粉通过影响雌激素和Hepcidin-FPN轴对肝脏铁代谢产生影响,维持部分铁稳态。 Objective To study the effects of long-term administration of ginseng powder on iron metabolism in serum and liver of ovariectomized rats.Methods ovariectomized rats were fed with 0.5,1and 3.0g/kg BWD ginseng powder mixed with basic diet respectively for 90days.Sham operation group and ovariectomized rats control group were given basic diet only;At the end of the experiment,serum estrogen and serum,liver iron metabolism related indicators were measured;The uterus was examined by gross and histopathology.Results compared with the sham operation group,the estrogen level(201.93±19.17vs 148.51±29.75)and the ratio of uterus to body(9.628±2.695vs 1.960±0.786)in the ovariectomized control group were significantly lower(P<0.01);The serum estrogen of ovariectomized rats treated with different doses of ginseng powder increased with the increase of ginseng dose(144.49±35.35vs 167.81±20.59vs 183.18±21.49),and the degree of uterine atrophy decreased gradually(1.952±0.527vs 3.120±1.120vs 4.379±1.602),and the difference between high-dose ginseng powder group and ovariectomized control group was statistically significant(P<0.05).There was no significant change in serum iron metabolism in each group;After ovariectomy,hepcidin mRNA expression level was significantly decreased(11.11±2.34vs 8.66±2.14),FPN mRNA expression level was increased(2.78±0.49vs 4.27±0.74),and BMP6mRNA expression level was decreased(4.56±0.44vs 3.93±0.73),which were significantly different from those in sham operation group(P<0.05);The remission of hepcidin in high-dose group was significantly higher than that in ovariectomized group(10.82±2.40,P<0.05),and the remission of FPN mRNA in high-dose and middle dose groups was significantly lower than that in ovariectomized control group(P<0.05);There was no significant effect of ginseng powder on BMP6mRNA expression.Conclusion a certain dose of ginseng powder can partially alleviate the adverse effects caused by the decrease of estrogen by regulating hepcidin,FPN mRNA expression levels in liver and serum estrogen levels in ovariectomized rats,suggesting thatginseng powder can affect liver iron metabolism and maintain partial iron homeostasis by affecting estrogen and hepcidin-FPN axis.
