摘要
枯草芽孢杆菌SX3411菌株自身带有subtilomycin基因簇,其主要的抑菌成分为subtilomycin.该文采用单因素分析法分析了枯草芽孢杆菌SX3411在不同发酵条件和营养成分时对抑菌物质产生的影响.根据单因素分析,对枯草芽孢杆菌SX3411产抑菌物质设计了响应面法分析,通过响应面法获得了最优的发酵条件和最佳营养成分配比.结果表明:最佳培养条件是初始pH值为8.25、转速为169 r·min^(-1)、温度为30.24℃、装液量为37.9%、接种量为1.46%,抑菌圈直径预测值为30.04 mm;最佳培养基的成分为葡萄糖11.4 g·L^(-1)、酵母粉5.0 g·L^(-1)、ZnSO_(4)0.00056 g·L^(-1)、CaCl_(2)0.0595 g·L^(-1),抑菌圈直径预测值为27.83 mm.实际检测值与预测值完全吻合.实验结果为生产羊毛硫细菌素subtilomycin提供了依据.
Bacillus subtilis SX3411 strain has subtilomycin gene cluster,and its main antimicrobial component is subtilomycin.The effects of Bacillus subtilis SX3411 on the production of antimicrobial substances under different fermentation conditions and nutritional components are analyzed by single factor method.According to the single factor analysis,the response surface methodology is designed to analyze the bacteriostatic substances produced by Bacillus subtilis SX3411.The optimal fermentation conditions and the optimal nutrient composition are obtained by response surface methodology.The results show that the optimum culture conditions are initial pH 8.25,rotational speed 169 r·min^(-1),temperature 30.24℃,liquid loading 37.9%,inoculation quantity 1.46%,and the predicted value of bacteriostasis circle diameter is 30.04 mm.The optimum medium is glucose 11.4 g·L^(-1),yeast powder 5.0 g·L^(-1),zinc sulfate 0.00056 g·L^(-1),calcium chloride 0.0595 g·L^(-1),and the predicted value of bacteriostasis circle diameter is 27.83 mm.The actual measured value is in good agreement with the predicted value.This provides a basis for the production of lantibiotic subtilomycin.
