薏苡仁细粉、破壁粉及破壁饮片物理指纹图谱和体外溶出度比较 被引量:1

Comparative Study of Physical Fingerprint and in vitro Dissolution of Coicis Semen Fine Powder,Broken Powder and Broken Decoction

摘要 目的比较薏苡仁细粉、破壁粉及破壁饮片的物理指纹图谱和体外溶出度。方法以薏苡仁为模型药物,建立其细粉、破壁粉及破壁饮片的物理指纹图谱,包括11个物理指标(指标松密度、振实密度、D_(90)、粒径分布宽度、粒径分布范围、豪斯纳比、休止角、颗粒间孔隙率、卡尔指数、吸湿性和含水率),并将其粉体物理属性归纳为堆积性、均一性、流动性、可压性和稳定性5个方面,采用夹角余弦法评价三者间的相似度;测定细粉、破壁粉及破壁饮片中多糖的溶出量和溶出速率,建立溶出动力学模型。结果薏苡仁细粉、破壁粉及破壁饮片均具有良好的堆积性、均一性、流动性和稳定性,细粉经超微粉碎后其堆积性、流动性略有降低,但经成型后得到了有效改善;由相似度可知,细粉、破壁粉及破壁饮片的物理指纹图谱相似度极高(>0.97);同时其溶出量、溶出速率大小排序为细粉<破壁粉<破壁饮片,溶出动力学模型均符合Weibull方程且溶出参数T30、T50、Td的大小排序为细粉>破壁粉>破壁饮片。结论通过物理指纹图谱构建和体外溶出度两方面的比较,薏苡仁破壁饮片具有明显优势。 Objective To compare the physical fingerprints and in vitro dissolution rates of Coicis semen fine powder,broken powder and broken decoction.Methods Taking Coicis semen as a model drug,the physical fingerprints of fine powder,broken powder and broken decoction are established,including 11 physical indexes(bulk density,vibration density,D_(90),diameter distribution width,diameter distribution range,hausnaby,angle of rerest,intergranular porosity,karl index,hygroscopicity and water content).The physical properties of the powder are summarized into five aspects,including stacking,homogeneity,fluidity,compressibility and stability,and the similarity among the three is evaluated by included angle cosine method;In addition,the dissolution rate and dissolution rate of polysaccharides in fine powder,broken wall powder and broken wall pieces are determined,and the dissolution kinetic model is established.Results The fine powder,broken powder and broken decoction of Coicis semen have good accumulation,uniformity,fluidity and stability.The accumulation and fluidity of fine powder decreased slightly after superfine grinding,but improved effectively after molding.At the same time,the order of dissolution amount and dissolution rate is fine powder<broken powder<broken decoction.The dissolution kinetic model conforms to Weibull equation,and the dissolution parameters T30,T50 and Td is fine powder>broken powder>broken decoction.Conclusion In this study,by comparing the construction of physical fingerprint and the dissolution in vitro,the Coicis semen broken decoction have obvious advantages.
