1株野生平菇菌株的鉴定及交配型分析 被引量:1

摘要 以1株野生平菇菌株为研究对象,采用ITS序列对其进行了种属鉴定,进一步通过单孢分离法获得了孢子单核菌株,经标准测交法和核迁移试验等对该平菇的交配型进行鉴定。结果表明,该野生平菇经ITS序列鉴定为糙皮侧耳,获得的69株孢子单核菌株,通过交配型鉴定得到T_(1)型20株、T_(2)型37株、T_(3)型9株、T_(4)型3株,通过核迁移试验鉴定T_(1)、T_(2)、T_(3)和T_(4)的交配型分别为A_(1)B_(2)_(1)、A_(2)B_(2)、A_(1)B_(2)和A_(2)B_(2)_(1),属于四极性异宗结合真菌,χ^(2)检测出现偏分离现象。 A strain of wild oyster mushroom was identified by ITS sequence analysis.The mating type of the wild strain was accurately identified by single spore isolation,standard cross test and nuclear migration test.The results showed that the wild oyster mushroom was identified as Pleurotus ostreatus.The 69 monokaryon strains obtained were identified as T_(1)(20 strains),T_(2)(37 strains),T_(3)(9 strains)and T_(4)(3 strains)by mating type identification.The mating types of T_(1),T_(2),T_(3)and T_(4)were identified as A_(1)B_(1),A_(2)B_(2),A_(1)B_(2)and A_(2)B_(1) respectively by nuclear migration test.It was proved that the wild oyster mushroom was a quaternary heterozygous combination fungus.The χ^(2) test showed segregation distortion rule.
