摘要
目的:探讨微小核糖核酸(miR)-612抑制白细胞分化抗原151(CD151)基因过表达对胃癌细胞侵袭和迁移能力的影响。方法:培养人胃癌细胞株SGC-7901,对其转染miR-612模拟物(miR-612组)或pcDNA3.1-CD151过表达载体(CD151组)后,实时荧光定量逆转录-聚合酶链反应(RT-qPCR)检测SGC-7901细胞中miR-612和CD151信使核糖核酸(mRNA)表达,用噻唑蓝(MTT)法、流式细胞仪和Transwell小室分别检测SGC-7901细胞增殖凋亡、侵袭迁移。结果:与miR-NC组比较,miR-612组细胞中miR-612表达水平明显升高,而CD151 mRNA表达水平明显降低(P<0.05)。与CD151组比较,在转染72 h后miR-612+CD151组细胞增殖活力明显降低(P<0.05),miR-612+CD151组细胞凋亡率明显高于CD151组(P<0.05)。Transwell小室检测结果显示,与对照组比较,miR-612组中侵袭细胞数和迁移细胞数均明显减少,而CD151组中侵袭细胞数和迁移细胞数均明显增多,差异均具有统计学意义(P<0.05);与CD151组比较,miR-612+CD151组中侵袭细胞数和迁移细胞数均明显减少,差异具有统计学意义(P<0.05)。结论:miR-612可通过靶向调控CD151表达抑制胃癌细胞增殖、侵袭迁移并促进细胞凋亡。
Objective To investigate the effect of miR-612 on gastric cancer cells by inhibiting CD151 gene overexpression invasion and migration.Methods Human gastric cancer cell line SGC-7901 was cultured and transfected with miR-612 mimicry(miR-612 group)or pcDNA3.1-CD151 overexpression vector(CD151 group).Then we detected the expression of miR-612 and CD151 mRNA in SGC-7901 cells by real-time quantitative reverse transcription-polymerase chain reaction(RT-qPCR).Proliferation,apoptosis,invasion and migration of SGC-7901 cells were detected by MTT method,flow cytometry and transwell chamber,respectively.Results Compared with the miR-NC group,the expression level of miR-612 in miR-612 group was significantly increased,while the expression level of CD151 mRNA was significantly decreased(P<0.05).Compared with CD151 group,the proliferation activity of miR-612+CD151 group was significantly decreased after transfection 72 h(P<0.05).The apoptosis rate of miR-612+CD151 group was significantly higher than that of CD151 group(P<0.05).Compared with the control group,the number of invading cells and migrating cells in Mir-612 group were decreased,while the number of invading cells and migrating cells in CD151 group increased significantly(P<0.05).The number of invading cells and migrating cells in Mir-612+CD151 group were decreased(P<0.05).Conclusion miR-612 can inhibit the proliferation,invasion,migration of SGC-7901 cells and promote apoptosis of gastric cancer cells by targeting CD151 expression.
