高通量芯片检测SLE家系成员自身抗体的临床价值探讨 被引量:1

摘要 目的探讨采用高通量芯片检测系统性红斑狼疮(SLE)家系成员的自身抗体的价值。方法选择该院确诊的7例SLE患者(来自3个SLE家系)作为病例组,家系1中未诊断为SLE的4名健康者作为对照组。通过高通量芯片法检测病例组和对照组自身抗体水平,比较检测结果的差异。结果热图结果显示,病例组120种自身抗体水平总体较对照组上调。与对照组比较,病例组抗dsDNA抗体、抗ssDNA抗体、抗染色体DNA抗体、抗胶原Ⅵ抗体、抗硫酸软骨素C抗体、抗GP210抗体、抗U1-snRNP 68/70抗体、抗U1-snRNP C抗体水平升高(P<0.05)。结论SLE家系自身抗体水平总体高于对照组,不同家系间表现出自身抗体的异质性,基于SLE家系筛选出自身抗体可为SLE诊断和精准治疗提供参考。 Objective To investigate the value of autoantibody detection by using high-throughput chip in systemic lupus erythematosus(SLE)family members.Methods Seven SLE patients(from three SLE families)diagnosed in this hospital were selected as the case group,and four healthy individuals who were not diagnosed with SLE in family 1 were selected as the control group.The autoantibodies levels in the case group and the control group were detected by using high-throughput chip method,and detection results were compared between groups.Results The heatmap results showed that the levels of 120 autoantibodies in the case group were higher than those in the control group.Compared with the control group,the levels of anti-dsDNA antibody,anti-ssDNA antibody,anti-chromosomal DNA antibody,anti-collagenⅥantibody,anti-chondroitin sulfate C antibody,anti-GP210 antibody,anti-U1-snRNP 68/70 antibody and anti-U1-snRNP C antibody in the case group increased(P<0.05).Conclusion The level of autoantibodies in SLE families is generally higher than that in the control group,and there is heterogeneity of autoantibodies among different families.Screening of autoantibodies based on SLE families can provide a reference for the diagnosis and precise treatment of SLE.
