摘要
目的:检测黑色素瘤相关抗原D4(MAGE-D4)在非小细胞肺癌(NSCLC)肿瘤组织及癌旁正常组织中的表达,探讨MAGE-D4在NSCLC组织中的表达及其与临床病理因素之间的关系。方法:收集2017年1月—2020年12月河南大学淮河医院病理科存档的242例NSCLC肿瘤组织蜡块及相应101例癌旁正常组织蜡块制成组织芯片。通过免疫组织化学法(SP法)检测MAGE-D4在242例NSCLC肿瘤组织及101例癌旁正常组织中的表达。结果:去除切片时损坏及漏切的肿瘤5例及周边组织1例,剩余肿瘤组织237例,癌旁正常组织100例的样本进行统计。MAGE-D4在肺癌组织中的表达阳性率为80.2%,明显高于癌旁组织49.0%(P<0.05)。通过χ^(2)检验得出MAGE-D4阳性表达与患者的年龄、肿瘤的大小、分期及分化程度呈正相关(P<0.05),而与性别、有无肺被膜侵犯、淋巴结转移及家族史、吸烟史等无明显关联性(P>0.05)。结论:非小细胞肺癌中存在MAGE-D4的异常表达,MAGE-D4阳性可能提示患者预后差。
Objective:To investigate the expression of melanoma associated antigen D4(MAGE-D4)in non-small cell lung cancer(NSCLC)tumor tissues and adjacent normal tissues,and to explore the relationship between MAGE-D4 expression in NSCLC tissues and clinicopathological factors.Methods:Tissue microarray was collected from 242 cases of NSCLC tumor tissue wax blocks and 101 cases of adjacent normal tissue wax blocks in the department of Pathology,Huaihe Hospital of He’nan University from January 2017 to December 2020.The expression of MAGE-D4 in 242 NSCLC tumor tissues and 101 adjacent normal tissues was detected by immunohistochemistry method(SP method).Results:When removing the section,there were 5 cases of damaged and missed tumor,1 case of surrounding tissue,237 cases of residual tumor tissue and 100 cases of paracancerous normal tissue.The positive rate of MAGE-D4 in lung cancer tissues was 80.2%which was significantly higher than that in paracancer tissues 49.0%(P<0.05).χ^(2) test showed that MAGE-D4 positive expression was positively correlated with age,tumor size,stage and differentiation degree(P<0.05),but had no significant correlation with gender,invasion of pulmonary capsule,lymph node metastasis,family history,smoking history,etc.(P>0.05).Conclusion:Abnormal expression of MAGE-D4 is found in NSCLC,and positive expression of MAGE-D4 may indicate poor prognosis.
