miR-204和Six1在甲状腺乳头状癌组织中的表达及意义 被引量:2

摘要 目的:研究甲状腺乳头状癌(PTC)组织中微小RNA-204(miR-204)、同源异型盒基因1(Six1)表达水平,并分析两者对PTC的诊断价值。方法:选取2016年3月—2019年3月湖北省枣阳市第一人民医院行手术治疗的PTC患者80例,取其手术切除的完整癌组织标本为PTC组,取对应癌旁组织为癌旁组。采用免疫组织化学法测定Six1蛋白表达情况;采用实时荧光定量PCR(RT-qPCR)法检测组织中miR-204、Six1 mRNA水平;Pearson法分析PTC组织中miR-204、Six1 mRNA水平的相关性;ROC曲线分析miR-204、Six1mRNA表达水平对PTC的诊断价值;分析miR-204、Six1蛋白表达与PTC临床病理因素的关系。结果:与癌旁组比较,PTC组miR-204/U6水平显著降低(P<0.05),Six1 mRNA水平及Six1蛋白阳性表达率显著升高(P<0.05)。Pearson分析结果显示,PTC组织中miR-204和Six1 mRNA水平呈负相关(r=-0.518,P<0.05)。ROC曲线分析显示,miR-204、Six1 mRNA水平诊断PTC的AUC分别为0.869、0.858,两者联合诊断的AUC为0.951。miR-204低表达、Six1蛋白呈阳性表达与肿瘤分化程度低、TNM分期为Ⅲ~Ⅳ期、淋巴结转移相关(P<0.05)。结论:PTC组织中miR-204异常低表达、Six1 mRNA异常高表达,两者联合检测对PTC有较高诊断价值。 Objective: To study the expression levels of microRNA-204(miR-204) and secreted in xylem 1(Six1) in papillary thyroid carcinoma(PTC) tissues, and to analyze their diagnostic value for PTC. Methods: A total of 80 patients with PTC who underwent surgical treatment at the First People’s Hospital of Zaoyang City from March 2016 to March 2019 were selected, the intact tumor tissue specimens were taken as PTC group, and the corresponding paracancerous tissue was taken as paracancerous group. The expression of Six1 protein was detected by immunohistochemistry;the levels of miR-204 and Six1 mRNA were detected by real-time quantitative PCR(RT-q PCR);Pearson method was used to analyze the correlation between miR-204 and Six1 mRNA levels in PTC patients;ROC curve was used to analyze the diagnostic value of miR-204 and Six1 mRNA expression levels in PTC;and the relationship between the expression of miR-204 and Six1 proteins and the clinicopathological parameters of PTC patients was analyzed. Results: Compared with those in the paracancerous group, the level of miR-204/U6 in PTC group was significantly lower(P<0.05), and the level of Six1 mRNA and positive expression rate of Six1 protein was significantly higher(P<0.05). Pearson analysis showed that miR-204 and Six1 mRNA levels were negatively correlated(r=-0.518, P<0.05). ROC curve analysis showed that the AUC of miR-204 and Six1 mRNA levels in the diagnosis of PTC were 0.869 and 0.858, respectively, and the AUC of the combined diagnosis of PTC was 0.951. Low expression of miR-204 and positive expression of Six1 protein were associated with low differentiation, TNM stage was III-IV and lymph node metastasis(P<0.05).Conclusions: The expression of miR-204 is abnormally low and the expression of Six1 mRNA is abnormally high in PTC. The combined detection of the two has high diagnostic value for PTC.
