摘要
目的:探究胃癌组织中长链非编码RNA-尿路上皮癌相关基因1(lncRNA UBC1)、骨形态发生蛋白2(BMP2)表达水平及其与临床病理特征、预后的关系。方法:选取2013年1月—2015年1月收治的胃癌患者79例作为研究对象,于术中收集胃癌组织及癌旁正常组织(距病灶>5 cm)。采用实时荧光定量PCR(qRT-PCR)法进行lncRNA UBC1、BMP2 mRNA表达水平检测,采用免疫组织化学染色法进行BMP2蛋白水平检测;采用Kaplan-Meier法分析不同lncRNA UBC1、BMP2蛋白水平患者生存情况;采用Cox回归模型分析胃癌患者预后不良的影响因素。结果:胃癌组织中lncRNA UBC1、BMP2 mRNA表达水平及BMP2蛋白高表达率均明显高于癌旁组织(P<0.05)。T_(3)~T_(4)、Ⅲ~Ⅳ期、淋巴转移患者胃癌组织中lncRNA UBC1、BMP2蛋白高表达率明显高于T_(1)~T_(2)、Ⅰ~Ⅱ期、无淋巴转移患者(P<0.05),且低分化胃癌组织中lncRNA UBC1、BMP2蛋白高表达率明显高于中、高分化胃癌组织,差异有统计学意义(P<0.05)。lncRNA UBC1、BMP2蛋白高表达患者5年累积生存率均明显低于lncRNA UBC1、BMP2蛋白低表达患者(P<0.05)。浸润深度(T_(3)~T_(4))、TNM分期(Ⅲ~Ⅳ期)、淋巴转移、lncRNA UBC1高表达、BMP2 mRNA高表达是胃癌患者不良预后发生的危险因素(P<0.05)。结论:胃癌组织中lncRNA UBC1、BMP2表达水平均明显升高,与分化程度、浸润深度、临床分期、淋巴转移等临床病理特征和患者预后不良发生密切相关。
Objective: To investigate the expression levels of long non-coding RNA urothelial bladder cancer associated transcript 1(lncRNA UBC1) and(BMP2) in gastric cancer and their relationship with clinicopathological features and prognosis. Methods: A total of 79 cases of gastric cancer patients in our hospital from January 2013 to January 2015 were selected as the research objects,and the gastric cancer tissues and adjacent normal tissues(more than 5 cm away from the lesion)were collected during the operation. Real-time fluorescence quantitative PCR(q RT-PCR) was used to detect the expression levels of lncRNA UBC1 and BMP2 mRNA, the protein level of BMP2 was detected by immunohistochemistry;Kaplan-Meier method was used to analyze the survival of patients with different levels of lncRNA UBC1 and BMP2;and Cox regression model was used to analyze the influencing factors of poor prognosis in patients with gastric cancer. Results: The expression levels of lncRNA UBC1, BMP2 mRNA and high expression rate of BMP2 protein in gastric cancer tissues were significantly higher than those in adjacent tissues(P<0.05). The high expression rates of lncRNA UBC1 and BMP2 protein in T_(3)-T_(4), Ⅲ-Ⅳ and lymph node metastasis patients were significantly higher than those in T_(1)-T_(2), Ⅰ-Ⅱ and non lymph node metastasis patients(P<0.05), the high expression rate of lncRNA UBC1 and BMP2 protein in poorly differentiated gastric cancer tissues is significantly higher than that in medium and well-differentiated gastric cancer tissues were statistically significant(P<0.05). The 5-year cumulative survival rate of patients with high expressions of lncRNA UBC1 and BMP2 was significantly lower than that of patients with low expressions of lnc RNA UBC1 and BMP2(P<0.05). The invasion depth of T_(3)-T_(4), TNM stage of stage Ⅲ-Ⅳ, lymph node metastasis, high expression of lncRNA UBC1 and high expression of BMP2 mRNA were the risk factors of poor prognosis in patients with gastric cancer(P<0.05). Conclusions: The expression levels of lncRNA UBC1 and BMP2 in gastric cancer tissues are significantly increased, which are closely related to the degree of differentiation, depth of invasion, clinical stage, lymph node metastasis and other clinicopathological features and poor prognosis of patients.
