The effects of LncRNA AWPPH on bone repair in periapical periodontitis

目的:研究LncRNA AWPPH在慢性根尖周炎中对骨修复的作用。方法:纳入慢性根尖周炎患者15例及牙周健康拔牙患者(对照组)10例,收集牙周组织样本,检测其中AWPPH的表达。对成骨细胞系MC3T3-E1转染AWPPH过表达载体,检测过表达AWPPH后细胞中BMP-2及RUNX-2的表达情况以及成骨细胞的增殖能力与分化能力。结果:AWPPH在慢性根尖周炎组织中表达低于对照组(P<0.05)。过表达AWPPH后BMP-2及RUNX-2的表达增高(P<0.05);过表达AWPPH后成骨细胞的增殖与分化能力增强(P<0.05)。结论:AWPPH在慢性根尖周炎中表达降低,过表达AWPPH可以促进成骨细胞增殖及分化。

Objective:To study the effects of AWPPH in chronic apical periodontitis.Methods:The expression of AWPPH in the samples of chronic periapical periodontitis(n=15)and the sumples of periapical healthy(control,n=10)was examined by qPCR.AWPPH was overexpressed in MC3T3-E1 cells by plasmid transfection,the expression of BMP-2 and RUNX-2 in the cells after overexpression of AWPPH was examined by qPCR and Western blot,the proliferation and differentiation ability of the cells were teste by CCK8 assay and ALP staining respectively.Results:The expression of AWPPH in chronic periapical periodontitis group was lower than that in the control group(P<0.05).After overexpression of AWPPH,the expression of BMP-2 and RUNX-2 in MC3T3-E1 cells increased(P<0.05);the proliferation and differentiation ability of the cells increased(P<0.05).Conclusion:The expression of AWPPH is decreased in chronic periapical periodontitis,over expression of AWPPH may promote the proliferation and differentiation of osteoblasts.
