摘要
目的探讨补肾活血方对DKK1阻断Wnt信号通路后薄型子宫内膜容受性相关蛋白表达的影响。方法建立雌性SD大鼠肾虚血瘀薄型子宫内膜联合模型,将SD大鼠分为对照组、模型组、补佳乐组、补肾活血方高剂量组、补肾活血方中剂量组、补肾活血方低剂量组、Wnt信号通路阻滞剂Dickkopf-1(DKK1)组、DKK1+中药组;各组给予相应的方法干预后,经过大鼠3个动情周期,处死取材。分别检测各组子宫内膜β连环蛋白(β-catenin)、OCT4基因及ABCG2蛋白的表达。结果补肾活血方高剂量组与补佳乐组比较,β-catenin、OCT4基因、ABCG2蛋白的表达差异无统计学意义(P>0.05);DKK1组与模型组比较,差异具有统计学意义(P<0.05);DKK1+中药组与DKK1组比较,差异具有统计学意义(P<0.01)。结论在改善β-catenin、OCT4基因、ABCG2蛋白表达方面,补肾活血方高剂量组能够达到与补佳乐相同的效果;补肾活血方可以增加DKK1阻断Wnt通路后β-catenin、OCT4基因、ABCG2蛋白的表达。
Objective To investigate the effect of Bushen Huoxue Recipe on thin endometrial tolerance-related proteins after DKK1 blocks the Wnt signaling pathway. Methods Female SD rats were divided into control group, model group, Bujiale group, Bushen Huoxue Recipe high-dose group, Bushen Huoxue Recipe medium-dose group, Bushen Huoxue Recipe low-dose group, Wnt signaling pathway blocker Dickkopf-1(DKK1) group and DKK1+ Chinese medicine group. Each group was given the corresponding method of intervention and then the rats were killed and extracted after 3 estrus cycles. The expression of β-catenin, OCT4 gene and ABCG2 protein in endometrium of each group were detected. Results There was no significant difference in the expression of β-catenin, OCT4 gene and ABCG2 protein between Bushen Huoxue Recipe high-dose group and Bujiale group(P>0.05). There were statistically significant differences between DKK1 group and model group(P<0.05). The difference between DKK1+ Chinese medicine group and DKK1 group was statistically significant(P<0.01). Conclusions In improving the expression of β-catenin, OCT4 gene and ABCG2 protein, Bushen Huoxue Recipe high-dose group could achieve the same effect as Bujiale;Bushen Huoxue Recipe can increase the expression of β-catenin, OCT4 gene and ABCG2 protein after DKK1 blocks Wnt pathway.
