黄酮类化合物通过炎症途径对心肌细胞的保护作用及机制的研究进展 被引量:8

摘要 黄酮类化合物在自然界中广泛分布,主要有抗炎、抗氧化、保护心血管等作用。心血管疾病严重威胁着人类健康,而炎症是引起心肌细胞损伤导致心血管疾病的关键因素。炎症在参与心肌细胞的氧化应激、凋亡等损伤过程中受到多种信号通路的共同调控,主要分为胞外的分泌型信号(细胞因子)、胞内的核转录因子-κB(nuclear transcription factor-κB,NF-κB)和丝裂原活化蛋白激酶(mitogen-activated protein kinase,MAPK)信号通路。本文通过对近年来相关文献整理归纳,总结出黄酮类化合物可通过降低肿瘤坏死因子-α(tumor necrosis factor-α, TNF-α)、白细胞介素-1(interleukin 1,IL-1)、IL-6等炎症细胞因子水平,抑制NF-κB和MAPK信号通路等炎症途径以减轻心肌细胞损伤、坏死和凋亡,从而发挥减轻心脏毒性和改善心脏功能等保护作用。其中,与心肌细胞炎症相关的信号通路还有Janus激酶2(Janus kinase 2,JAK2)/转录激活蛋白3 (signal transducerand activator of transcription 3,STAT3)、Toll样受体4(toll-like receptor 4,TLR4)和磷脂酰肌醇3-激酶(phosphatidylinositol 3-kinase,PI3K)/蛋白激酶B(protein kinase B,Akt)信号通路。此外,在炎症反应过程中黄酮类化合物具有抑制氧化应激的保护作用。本文综述了黄酮类化合物通过炎症途径对心肌细胞的保护作用及机制的研究进展,以期为开发防治心血管疾病药物提供新的研究思路。 Flavonoids are widely distributed in the nature,which have the effects of antiinflammatory,anti-oxidation,cardiovascular protection and others.Cardiovascular disease is a serious threat to human health.Inflammation is the key factor leading to myocardial cell damage and cardiovascular disease.Inflammation is regulated by a variety of signal pathways in the process of oxidative stress,apoptosis and other injury of myocardial cells,which are mainly divided into extracellular secretory signaling(cytokines),intracellular nuclear transcription factors-κB(NF-κB)and mitogen-activated protein kinase(MAPK)signal pathway.Based on the relevant literature in recent years,it is concluded that flavonoids can reduce tumor necrosis factor by reducing tumor necrosis factor-α(TNF-α)、interleukin-1(IL-1),IL-6 and other inflammatory cytokines level,inhibiting inflammatory pathways such as NF-κB and MAPK signal pathway,which can reduce the injury,necrosis and apoptosis of myocardial cells,thus play a protective role in reducing cardiac toxicity and improving cardiac function.Among them,Janus kinase 2(JAK2)/signal transducer and activator of transcription 3(STAT3),toll like receptor 4(TLR4)and phosphatidylinositol 3-kinase(PI3K)/protein kinase B(Akt)signal pathways are also involved in myocardial inflammation.In addition,flavonoids can inhibit oxidative stress in the process of inflammation.In this paper,we reviewed the research progress on the protective effect and mechanism of flavonoids on the cardiomyocyte through inflammatory pathway,in order to provide new research ideas for the development of drugs for the prevention and treatment of cardiovascular diseases.
