摘要
目的鉴定马尔尼菲篮状菌(TM)的分泌蛋白,并初步分析其生物学功能。方法基于质谱技术对TM菌株(ATCC18224)培养上清进行分泌蛋白组鉴定,利用生物信息学手段预测所鉴定蛋白的亚细胞定位和信号肽剪切位点,以及分析其生物学功能和信号通路。结果通过2次独立重复实验,共鉴定得到21个分泌蛋白;功能注释结果显示分泌蛋白主要参与超氧化物代谢、压力应对、金属离子转运等生物学过程;预测结果显示,分泌蛋白主要定位于细胞核、细胞外、质膜、线粒体和细胞膜,且仅有6个蛋白存在信号肽序列。结论TM分泌蛋白可能与其免疫逃逸、真菌毒性密切相关,且部分蛋白功能尚不明确,需要深入研究。
The purpose of this research was to identify Talaromyces marneffei(TM)exported proteins and analyze their biological functions.With mass spectrometry technology,culture supernatants from the TM strain(ATCC18224)were used to identify exported proteins,and bioinformatics methods were used to predict the subcellular localization and signal peptide cleavage sites of the identified proteins,as well as to analyze their biological functions and signaling pathways.Through two independent repeated experiments,a total of 21 secreted proteins were identified.The functional annotation results indicated that the exported proteins are mainly involved in biological processes such as superoxide metabolism,stress response,and metal ion transport;the prediction results showed that the exported proteins are mainly located in the nucleus,extracellular,plasma membrane,mitochondria,and cell membranes.Only six proteins had signal peptide sequences.In summary,TM exported proteins may be closely associated with TM immune escape and fungal toxicity.However,the functions of some proteins remain unclear and require in-depth study.
