睡眠方颗粒剂对对氯苯丙氨酸致失眠大鼠单胺类神经递质的影响 被引量:2

Effect of sleep formula granules on levels of monoamine neurotransmitters in insomnia rats induced by DL-4-Chlorophenylalanine

摘要 目的研究睡眠方颗粒剂镇静催眠作用,初步探讨其作用机制。方法将60只SD大鼠随机分为正常组、模型组、阳性对照药睡眠方丸剂组和睡眠方颗粒剂高剂量组、中剂量组、低剂量组,每组10只大鼠。除正常组外,其余各组大鼠均建立失眠模型。模型成功后,睡眠方丸剂组和睡眠方颗粒剂高、中、低剂量组分别给予相应的药物灌胃,模型组和正常组均给予同等体积的0.9%生理盐水灌胃。比较各组大鼠脑组织5-羟色胺(5-HT)、5-羟吲哚乙酸(5-HIAA)、多巴胺(DA)和去甲肾上腺素(NA)水平。结果与模型组比较,睡眠方组大鼠脑组织5-HT、5-HIAA含量升高(P<0.05),DA、NA含量降低(P<0.05);与睡眠方丸剂组比较,睡眠方颗粒剂高剂量组大鼠脑组织5-HT、5-HIAA含量升高(P<0.05),NA含量降低(P<0.05),睡眠方颗粒剂中剂量组差异无统计学意义(P>0.05)。结论睡眠方颗粒剂有改善大鼠失眠的作用,高、中剂量改善失眠大鼠作用强于低剂量组。对5-HT、5-HIAA、DA和NA的调节作用可能是其治疗失眠的作用机制。 Objective To investigate the sedative and hypnotic effects and mechanisms of sleep formula granules.Methods A total of 60 adult Wistar rats were divided into normal group,model group,sleep formula pill group and sleep formula granule high dose group,medium dose group,low dose group and 10 rats each group.The insomnia model was established in 5 groups except for the control group.After the success of the model,sleep formula pill group and sleep formula granule high dose group,medium dose group,low dose group were given corresponding drug gavage,Model group and normal group were given the same volume of 0.9% normal saline gavage.The 5-HT,5-HIAA,NA and DA contents in brain tissue were detected and compared.Results Compared with the model group,the content of 5-HT and 5-HIAA in brain tissue of rats in sleep formula granule were increased(P<0.05),while the content of DA and NA were decreased(P<0.05).Compared with the sleep formula pill group,the content of 5-HT and 5-HIAA in brain tissue of rats in high dose of sleep formula granule were increased(P<0.05),while the content of NA was decreased(P<0.05).There was no statistical difference between sleep formula pill group and medium dose of sleep formula granule(P>0.05).Conclusion Sleep formula granule can improve insomnia in rats.The high dose and medium dose of sleep formula granule has stronger effect than low dose,and the regulating effect on 5-HT,5-HIAA,NA and DA may be the mechanism of insomnia treatment.
