温肾健脾法对腹泻型肠易激综合征大鼠背根神经节NMDA受体及其磷酸化表达的影响 被引量:3

摘要 目的观察温肾健脾法对腹泻型肠易激综合征(Irritable Bowel Syndrome,IBS)模型大鼠背根神经节NMDA受体及其磷酸化表达的影响。方法参照课题组前期造模方法,采用母子分离结合番泻叶灌胃,将28只SPF级SD大鼠乳鼠及SD大鼠母鼠随机分为正常对照组、模型组、温肾健脾方组、匹维溴铵组共4组,每组7只。用Western Blot和免疫组化的方法检测各组大鼠背根神经节中NMDAR1、NMDAR2B及其磷酸化的表达。结果模型组背根神经节组织中NMDAR1、phospho-NMDAR1、NMDAR2B、phospho-NMDAR2B表达组间差异明显,模型组背根神经节组织中的NMDAR1、phospho-NMDAR1、NMDAR2B、phospho-NMDAR2B表达较正常对照组明显升高(P<0.01),药物干预后,温肾健脾方组和匹维溴铵组大鼠背根神经节组织中NMDAR1、phospho-NMDAR1、NMDAR2B、phospho-NMDAR2B的表达与模型组相比明显下调(P<0.01)。温肾健脾方组和匹维溴铵组两组间NMDAR1、phospho-NMDAR1、NMDAR2B、phospho-NMDAR2B的表达差异无统计学意义(P>0.05)。结论温肾健脾方通过调节腹泻型肠易激综合征模型大鼠背根神经节中的NMDA受体及其磷酸化表达可能是其降低内脏高敏感的机制之一。 Objective To observe the effect of the method of warming the kidney and strengthening the spleen on the NMDA receptor and its phosphorylation expression in the dorsal root ganglia of diarrheal irritable bowel syndrome(Irritable Bowel Syndrome,IBS)rats.Methods According to the previous modeling method of the research group,28 neonatal rats were divided into 4 groups randomly,including normal control group,model group,Wenshen Jianpi Decoction group and pivironium bromide group,with 7 rats in each group.The expressions of NMDAR1,NMDAR2 B and their phosphorylation in dorsal root ganglion of rats were detected by Western Blot and immunohistochemistry.Results The expression of NMDAR1,phospho-NMDAR1,NMDAR2 B,and phospho-NMDAR2 B in the dorsal root ganglion tissue of the model group was significantly different between the groups,and the expression of NMDAR1,phospho-NMDAR1,NMDAR2 B,and phospho-NMDAR2 B in the dorsal root ganglion tissue of the model group was relatively normal.The control group increased significantly(P<0.01).After drug intervention,the expression and model of NMDAR1,phospho-NMDAR1,NMDAR2 B,and phospho-NMDAR2 B in the dorsal root ganglion tissue of rats in the Wenshen Jianpi prescription group and pinaverium bromide group Compared with the group,it was significantly down-regulated(P<0.01).There was no significant difference in the expression of NMDAR1,phospho-NMDAR1,NMDAR2 B,and phospho-NMDAR2 B between the Wenshen Jianpi prescription group and the pinaverium bromide group(P>0.05).Conclusion The regulation of NMDA receptor and its phosphorylated expression in dorsal root ganglion of rats with diarrhoea-type irritable bowel syndrome may be one of the mechanisms of Wenshen Jianpi Decoction to reduce visceral hypersensitivity.
