温度响应性水凝胶的制备及其对姜黄素的控制释放研究 被引量:1

摘要 目的姜黄素是一种天然活性分子,其较好的抗炎、抗氧化作用,在伤口修复抗炎领域具有很好的应用前景,但是较差的水分散性限制了它的应用。本研究采用一锅法将N-异丙基丙烯酰胺(N-isopropylacrylamide,NIPAM)、接枝双键的Pluronic F-127(PF127-DA)、葡聚糖及双丙烯酰胺(Bis-acrylamide,BIS)混合,在过硫酸铵(Ammonium persulfate,APS)引发、四甲基乙二胺(N,N,N′,N′-Tetramethylethylenediamine,TEMED)催化下成胶,通过水凝胶负载姜黄素的方式大大扩大了姜黄素的应用场景。方法利用Pluronic F-127(PF-127)胶束包载姜黄素,聚-(N-异丙基丙烯酰胺)[Poly-(N-isopropylacrylamide),PNIPAM]温度响应释药。结果该水凝胶具有良好的温度响应性能,溶胀性能,均匀的孔结构以及良好的药物缓释性能。结论该新型水凝胶极大地提高了姜黄素的水溶性和达到对姜黄素的控制释放,将在温度响应载药水凝胶领域具有广阔的应用前景。 Objective Curcumin is a natural active molecule with good anti-inflammatory and antioxidant effects.It has a good application prospect in the field of wound repair and anti-inflammatory,but its poor water dispersion limits its application.In this study,N-isopropylacrylamide(NIPAM),Pluronic F-127(PF127-DA),dextran and BIS acrylamide(BIS)were mixed by one pot method.Tetramethylethylenediamine was initiated by ammonium persulfate(APS)(N,N,N′,N′Tetramethylethylenediamine,TEMED)catalyzed the formation of curcumin,which greatly expanded the application of curcumin through the way of hydrogel loading curcumin.Methods Curcumin was encapsulated in Pluronic F-127(PF-127)micelles [Poly-(N-isopropylacrylamide),PNIPAM]temperature responsive drug release.Results The hydrogel has good temperature response,swelling property,uniform pore structure and good drug release properties.Conclusion The new hydrogel greatly improves the water solubility of curcumin and the controlled release of curcumin.It will have broad application prospects in the field of temperature responsive hydrogel.
