白眉草总黄酮超声辅助提取工艺优化及抗菌活性研究 被引量:7

Optimization of ultrasonic-assisted extraction technology and antibacterial activity of total flavonoids from Gerbera piloselloides (L.) Cass

摘要 本文以白眉草为试验材料,采用超声辅助法优化白眉草总黄酮提取工艺,并研究其抗菌活性。采用单因素结合正交设计试验,考察了乙醇浓度、料液比、超声温度和超声时间对总黄酮得率的影响。结果表明:白眉草总黄酮最佳提取工艺条件为:乙醇浓度70%,料液比1:20,超声温度60℃,超声时间45 min。在最佳工艺下白眉草总黄酮得率为3.96%,工艺稳定,可行。白眉草总黄酮提取物对大肠杆菌和金黄色葡萄球菌均有较强的抑制作用,对大肠杆菌的最低抑菌浓度(MIC)为12.5 g/L,最低杀菌浓度(MBC)为25 g/L;对金黄色葡萄球菌的MIC为25 g/L,MBC为50 g/L。本研究为白眉草在饲料添加剂中的开发利用提供一定依据。 Taking Gerbera piloselloides(L.)Cass as raw material,the extract technology of total flavonoids was optimized with ultrasonic assisted method,and the antibacterial activity was studied.Based on the single factor experiment,the effect of ethanol concentration,material-liquid ratio,ultrasonic temperature and ultrasonic time on the yield of total flavonoids was tested with orthogonal design.The results showed that the optimal conditions of total flavonoids from Gerbera piloselloides were ethanol concentration of 70%,material-solid ratio of 1:20,ultrasonic temperature of 60℃and ultrasonic time of 45 min.Under the optimal conditions,the yield of total flavonoids was 3.96%.The total flavonoids from Gerbera piloselloides(L.)Cass had strong inhibitory effect on Escherichia coli and Staphylococcus aureus,the MIC to Escherichia coli was 12.5 g/L,the MBC to Escherichia coli was 25 g/L,and the MIC to Staphylococcus aureus was 25 g/L,the MBC to Staphylococcus aureus was 50 g/L.This study provide a theoretical basis for the further development and utilization in feed additive of the Gerbera piloselloides(L.)Cass.
