高效液相色谱法检测饲料中6种游离氨基酸

High performance liquid chromatography method for determination of six free amino acids in feed

本实验通过高效液相色谱法对饲料中6种游离氨基酸含量进行测定。采用0.1 mol/L盐酸溶液提取样品中的6种游离氨基酸,与1%2,4-二硝基氟苯(DNFB)溶液衍生后经高效液相色谱仪测定。结果表明:饲料中6种游离氨基酸平均回收率为97.52%,检出限≥0.006%(信噪比S/N=3),定量限≥0.021%(信噪比S/N=10),重复性以变异系数计≥2.55%。综上,该方法适合饲料中6种游离氨基酸的测定。

High performance liquid chromatography(HPLC)method for determination of six free amino acids in feed.Six free amino acids were extracted by 0.1mol/L hydrochloric acid solution,derivatived by 1%2,4-Dinitrofluorobenzene,and determined with HPLC.The results showed that the mean recoveries of feed was 97.52%.The limit of detection(LOD)≥0.006%(S/N=3)and the limit of quantitation(LOQ)≥0.021%(S/N=10).The repeatability was calculated as the coefficient of variation≥2.55%.This method could be used for quantitive analysis of six free amino acids in feed.
