摘要
目的高产庆大霉素产生菌的筛选。方法对出发菌株绛红小单孢菌GM20190109-15进行常温等离子体(ARTP)和氯化锂复合诱变处理。结果经高通量筛选,得到一株突变菌株CH20190225-107,经高效液相色谱检测,突变菌株的各组份出峰时间与对照一致,其效价从1547 U/mL提高到2280 U/mL,摇瓶效价较出发菌株提高了47.4%,组分C1、C1a、C2a和C2的相对比例分别为29%、23%、25%和23%,且遗传性状稳定。结论使用ARTP和氯化锂复合诱变庆大霉素产生菌的,效果较好,提高了庆大霉素的产量和质量。
Objective In order to obtain high-yield Micromonospora purprua and improve the productivity and quality of GM.Methods The original strain GM20190109-15 is multiple mutated by ARTP&lithium chloride.Result We obtain a mutant strain CH20190225-107 by high throughput screening.The peak time of each component of the mutant strain is consistent with the contrast by high performance liquid chromatography.The potency is increased from 1547 U/mL to 2280 U/mL.The shake flask titer increased by 47.4%compared with the original strain,the relative proportions of component C1,C1a,C2a,and C2 are 29%,23%,25%,and 23%respectively,as well as stable inheritable.Conclusion In this study,it is effective for improving the yield and quality of gentamicin by the combined mutagenesis of ARTP and lithium chloride.
