枳术丸对功能性消化不良的治疗作用机制探讨

Discussion on Therapeutic Mechanism of Zhizhu Pills on Functional Dyspepsia based on Network Pharmacology

目的:运用网络药理学技术探讨枳术丸对功能性消化不良的作用机制。方法:在TCMSP平台中检索出枳术丸的有关活性成分并进行初步筛选,借助R语言与Uniprot数据库找到活性成分对应的统一规范靶点,在GeneCards数据库中检索到功能性消化不良的作用靶点并进行初步筛选。将二者得到的靶点取交集得到共同靶点,把共同靶点导入String数据库得到PPI蛋白互作网络图。再在DAVID数据库中导入共同靶点信息进行GO分析和KEGG分析。结果:检索筛选得到枳术丸活性成分29种,对应靶点62个,功能性消化不良靶点553个,共同靶点23个。其中关键靶点有EGFR、IL6、VEGFA、CASP3,主要参与凋亡负调控、上皮细胞增殖、氨基酸反应、雌二醇反应、一氧化氮合成等生物进程,涉及的通路有癌症途径、HIF-1信号通路、TNF信号通路等。结论:枳术丸对功能性消化不良的治疗是多成分、多靶点、多途径共同作用的结果,有助于进一步阐释枳术丸的药理作用机制。

Objective:To explore the mechanism of Zhizhu pills on functional dyspepsia using network pharmacology technology.Method:Retrieve the relevant active ingredients of Zhizhu pills from the TCMSP platform and conduct a preliminary screening.Use the R language and Uniprot database to find the unified normative targets corresponding to the active ingredients,and retrieve the functional dyspepsia targets in the GeneCards database.And conduct a preliminary screening.Take the intersection of the two targets to obtain a common target,and import the common target into the String database to obtain the PPI protein interaction network diagram.Then import the common target information into the DAVID database for GO analysis and KEGG analysis.Results:29 active ingredients of Zhizhu pills were retrieved and screened,corresponding to 62 targets,553 functional dyspepsia targets,and 23 common targets.Among them,the key targets are EGFR,IL6,VEGFA,and CASP3,which are mainly involved in biological processes such as negative regulation of apoptosis,epithelial cell proliferation,amino acid response,estradiol response,and nitric oxide synthesis.The pathways involved include cancer pathway,HIF-1 Signal pathway,TNF signal pathway,etc.Conclusion:The treatment of Zhizhu pills for functional dyspepsia is the result of multiple components,multiple targets,and multiple pathways,which will help to further explain the pharmacological mechanism of Zhizhu pills.
