摘要
为了解我国东南沿海地区三带喙库蚊抗药性水平,在海南、福建、浙江和江苏等4个省份采集5个自然种群三带喙库蚊成蚊,采用美国CDC生测瓶法和分子生物学方法分别检测各种群三带喙库蚊成蚊对两种拟除虫菊酯类杀虫剂的敏感性和kdr等位基因频率。结果显示,海口美兰(HKML)、泉州南安(QZNA)、金华临海(JHLH)、台州浦江(TZPJ)和连云港板铺(LYGBP)等5个种群三带喙库蚊成蚊对0.05%溴氰菊酯的KT_(50)值在2.17~16.96 min之间,接触1 h后的击倒率除JHLH种群为98.39%以外,其余种群均为100%,24 h死亡率均为100%;对0.05%高效氯氰菊酯的KT_(50)值在3.90~16.23 min之间,接触1 h后的击倒率和24 h死亡率均为100%;各种群成蚊kdr等位基因频率在0~38.89%之间,kdr等位基因频率与其对溴氰菊酯和高效氯氰菊酯的KT_(50)值之间相关性显著,且均为正相关关系。根据kdr等位基因频率和生物测定结果,推测QZNA种群三带喙库蚊对溴氰菊酯和高效氯氰菊酯还处于相对敏感阶段,HKML种群已经产生了部分抗性,TZPJ、LYGBP和JHLH种群的抗性程度相对较高;在使用美国CDC生测瓶法对三带喙库蚊成蚊进行抗性检测时,0.05%溴氰菊酯(1 h)和0.05%高效氯氰菊酯(1 h)的诊断剂量偏高。本次研究结果为上述地区的三带喙库蚊成蚊防制和卫生杀虫剂使用策略提供了一定的科学依据。
To survey the resistance of Culex tritaeniorhynchus to pyrethroid insecticides in southeast coastal areas of China,adult samples of Cx.tritaeniorhynchus were collected from 5 sites in 4 provinces(Hainan,Fujian,Zhejiang and Jiangsu).The sensitivity of Cx.tritaeniorhynchus to two pyrethroid insecticides and kdr alleles frequencies were determined with CDC bottle bioassay and molecular biology method respectively.The results showed that the KT_(50)value of 0.05%deltamethrin in 5 Cx.tritaeniorhynchus populations(HKML,QZNA,JHLH,TZPJ and LYGBP)ranged from 2.17 to 16.96 min,the 1 h knockdown rate of JHLH strain exposed to deltamethrin was 98.39%,while that of the other four populations were 100%,and the 24 h mortalities of all populations were 100%.The KT_(50)value of 0.05%betacypermethrin ranged from 3.90 to 16.23 min,and both 1 h knockdown rate and 24 h mortalities rate of all populations were 100%.The kdr alleles frequencies ranged from 0 to 38.89%,and the kdr alleles frequency of Cx.tritaeniorhynchus was positively correlated with the KT_(50)values of deltamethrin and betacypermethrin.According to the kdr alleles frequencies and bioassay results,it was conjectured that the QZNA population was still at a relatively sensitive stage to deltamethrin and beta-cypermethrin,HKML population has been produced partial resistance,while the TZPJ,LYGBP and JHLH populations showed relatively higher resistance.The diagnostic doses of 0.05%deltamethrin(1 h)and 0.05%betacypermethrin(1 h)for adult Cx.tritaeniorhynchus were too high for using CDC bottle bioassay.The present results provided a certain scientific basis for adult mosquito control and hygienic insecticide use of Cx.tritaeniorhynchus in these mentioned above.
