维甲酸对异位子宫内膜组织自噬和血管生成的影响

Effects of Retinoic Acid on Autophagy and Angiogenesis in Ectopic Endometrial Tissue

摘要 目的:观察小鼠异位子宫内膜组织中自噬标记物Beclin 1及血管内皮生长因子(Vascular endothelial growth factor,VEGF)的表达变化情况,探讨维甲酸(Retinoic Acid,RA)对异位子宫内膜病灶中自噬及血管生成的作用。方法:100只BALB/c雌性小鼠分为5组:CTRL组,EMs组,LOW组[100 mg/(kg·d)],HIGH组[200 mg/(kg·d)],CQ组;药物灌胃7 d后处死取材,行Real-time PCR,Western blotting定量分析及MVD测定。结果:EMs组、LOW组Beclin 1、VEGF mRNA表达均高于CTRL组,差异均有统计学意义(P<0.05);HIGH组Beclin 1、VEGF mRNA表达均低于LOW组,差异均有统计学意义(P<0.05)。EMs组、LOW组Beclin 1、VEGF蛋白表达均高于CTRL组,差异均有统计学意义(P<0.05);HIGH组Beclin 1、VEGF蛋白表达均低于LOW组,差异均有统计学意义(P<0.05)。EMs组、LOW组MVD计数均高于CTRL组,差异均有统计学意义(P<0.05);HIGH组MVD计数低于LOW组和EMs组,差异均有统计学意义(P<0.05)。结论:高剂量维甲酸可抑制EMs异位病灶中的自噬及血管生成。 Objective:To observe the expression of autophagy marker Beclin 1 and vascular endothelial growth factor(VEGF)in mouse ectopic endometrium,and to explore the effect of Retinoic Acid(RA)on autophagy and angiogenesis in ectopic endometrium.Method:100 BALB/c female mice were divided into 5 groups,CTRL group,EMs group,LOW group[100 mg/(kg·d)],HIGH group[200 mg/(kg·d)],CQ group;after 7 days of drug gavage,they were killed and taken for real-time PCR,Western blotting quantitative analysis and MVD determination.Result:The mRNA expressions of Beclin 1 and VEGF in EMs group and LOW group were higher than those in CTRL group,the differences were statistically significant(P<0.05);the mRNA expressions of Beclin 1 and VEGF in HIGH group were lower than those in LOW group,the differences were statistically significant(P<0.05).The protein expressions of Beclin 1 and VEGF in EMs group and LOW group were higher than those in CTRL group,the differences were statistically significant(P<0.05);the protein expressions of Beclin 1 and VEGF in HIGH group were lower than those in LOW group,the differences were statistically significant(P<0.05).The MVD counts in EMs group and LOW group were higher than those in CTRL group,the differences were statistically significant(P<0.05);the MVD count in HIGH group was lower than that in LOW group and EMs group,the differences were statistically significant(P<0.05).Conclusion:High dose Retinoic Acid can inhibit autophagy and angiogenesis in ectopic lesions of EMs.
