Simultaneous Determination of 21 Active Components in Renshen(Panax Ginseng), Tianma(Gastrodia elata) and Their Compounds by HPLC

摘要 目的建立HPLC法同时测定人参、天麻及其复方中21种活性成分(天麻素、对羟基苯甲醇、人参皂苷-Rg_(1)、Re、Rg_(2)、Rg_(3)、Rg_(5)、Rf、F1、F2、Rc、Rd、Rb_(1)、Rb_(2)、Rb_(3)、Rh_(2)、人参皂苷C-K、20(R)-人参皂苷-Rh1、人参皂苷-Rh_(4)、原人参二醇及原人参三醇)的含量。方法采用Hypersil BDS C_(18)色谱柱(250 mm×4.6 mm,5μm),以乙腈(A)-水(B)为流动相;梯度洗脱;流速1.0 mL·min^(-1);检测波长203 nm;柱温35℃;进样量20μL。结果21种活性成分质量浓度分别在考察的线性范围内与峰面积呈良好的线性关系(r≥0.999)。加样回收率为97.71%~102.33%(RSD≤1.95%)。甲醇提取对人参天麻复方药效成分含量无显著影响,其含量较高的主要是天麻素,人参皂苷-Rg1、Re、Rb1、Rc、Rg2。结论建立的天麻人参复方中21种活性成分的含量测定方法简便、准确可靠,可以为人参、天麻及其复方制剂的质量控制提供新的高效的评价方法. Objective To establish an HPLC method for the simultaneous determination quantification of 21 active components[gastrodin,p-hydroxybenzyl alcohol,ginsenoside Rg_(1),Re,Rg_(2),Rg_(3),Rg_(5),Rf,F1,F2,Rc,Rd,Rb_(1),Rb_(2),Rb_(3),Rh_(2),Compound K,20(R)-Rh1,Rh_(4),protopanaxdiol and protopanaxatriol]in Renshen(Panax Ginseng),Tianma(Gastrodia elata)and their compounds.Methods The HPLC separation was performed on an Hypersil BDS C_(18) column(250 mm×4.6 mm,5μm)in a gradient elution mode with mobile phase consisting of methanol and water at a flow rate of 1.0 mL·min^(-1).The detection wavelength was set at 203 nm and the column temperature was 35℃.The injection volume was 20μL.Results There was a good linear relationship between the mass concentration of 21 active components and the peak area(r≥0.999).The recovery rates was 97.71%~102.33%(RSD≤1.95%).The methanol extraction had no significant effect on the contents of active components in Renshen(Panax Ginseng),Tianma(Gastrodia elata)before and after compatibility,and the contents of gastrodin,ginsenoside Rg_(1),Re,Rb_(1),Rc and Rg_(2) were higher.Conclusion The determination method for 21 active components in Renshen(Panax Ginseng),Tianma(Gastrodia elata)compound is simple,accurate and reliable,which provides a new and efficient technique for the quality control of Renshen(Panax Ginseng),Tianma(Gastrodia elata)and their compound preparations.
