高效液相色谱法测定蒙成药拉克都拉-12中栀子苷的含量

Determination of Gardenoside in Mongolian Medicine Lakedula-12 by HPLC

目的建立测定蒙成药拉克都拉-12中栀子苷含量的高效液相色谱法。方法以乙腈-水(15∶85)为流动相,流速1.0 ml/min,检测波长为238 nm,采用C18色谱柱,柱温30℃,进样量为10μl,外标法定量。结果栀子苷在0.0271~0.5413μg范围内与峰面积呈良好的线性关系(r=0.9999),平均回收率为91.2%(RSD=0.9%)。结论该测定方法快速、简便、专属性较强,可用于拉克都拉-12中栀子苷的含量测定和质量控制。

Objective To establish the determination of content of gardenoside in Mongolian medicine Lakedula-12 by high performance liquid chromatography.Methods Acetonitrile-water(15:85)was used as mobile phase at the flow rate of 1.0 ml/min.The detection wavelength was set at 238 nm.A C18 column was used and the temperature was 30℃.The sample volume was 10μL.Quantification was determined by external standard method.Results The linear relationship between gardenoside and peak area was good in the range of 0.0271-0.5413μg(r=0.9999).The average recovery was 91.2%(RSD=0.9%).Conclusion The method is rapid,simple and specific,and can be used for the content determination and quality control of gardenoside in Lakedula-12.
