摘要
为明确取食不同食物金针虫的肠道微生物群落结构差异以及寄主抵御外界不良因子干扰的影响,运用MiSeq测序技术比较分析平沙绿僵菌Metarhizium pingshaense处理取食小麦和马铃薯沟叩甲Pleonomus canaliculatus的肠道细菌群落结构差异。结果显示,不同处理沟叩甲种群之间Simpson指数差异显著,但Chao1、Observed species、PD whole tree和Shannon指数差异均不显著;肠道细菌类微生物主要由厚壁菌门、拟杆菌门、放线菌门和变形杆菌门组成,取食马铃薯种群、饥饿种群和平沙绿僵菌处理的饥饿种群厚壁菌门的相对含量分别为29.46%、39.46%和29.81%;对照种群、取食小麦种群和平沙绿僵菌处理的马铃薯种群拟杆菌门的相对含量分别为31.30%、29.79%和39.07%;取食小麦种群中普通拟杆菌Bacteroides vulgatus PC510优势菌群的相对含量最高达8.37%,其余处理种群中优势菌群相对含量均低于5.00%。对不同处理沟叩甲种群进行相关性分析,发现对照种群与平沙绿僵菌处理种群的相关性较低,但与其余处理组相关性均较高;PCA聚类结果显示,平沙绿僵菌处理种群与其余种群之间聚集度较低。表明平沙绿僵菌的胁迫对沟叩甲肠道内细菌微生物群落多样性、丰富度及结构变化存在一定程度的影响,且外源食物摄入与平沙绿僵菌的胁迫两者叠加效应对寄主昆虫肠道微生物群落结构的影响高于单一外源因素的影响。
MiSeq sequencing was used to evaluate the intestinal microbiome in gut of canaliculated wireworm Pleonomus canaliculatus with different feeding/starving treatments.The results showed that there was a significant difference in Simpson index,but no differences in index of Chao1,observed species,PD whole tree and Shannon index among different feeding/starving treatments.The gut bacteria of wireworm were mainly consisted of phyla Firmicutes,Bacteroidetes,Actinomycetes and Proteus.The relative abundance of Firmicutes in potato-fed,starvation,and Metarhizium pingshaense-treated starvation wireworm were 29.46%,39.46% and 29.81%,respectively.The Bacteroides proportions in control,wheat-fed,combination of M.pingshaense-treated and potato-fed populations were 31.30%,29.79%and 39.07%,respectively.The dominance of Bacteroides vulgatus PC510 in wheat-fed population was the highest(8.37%),whereas the dominant bacteria were less than 5%in other treatments.The correlation analysis showed that the control had high correlation to other treatments M.pingshaense treated populations.The aggregation degree of the M.anisopliae treated populations was lower than the other populations.This indicated that the stress of M.pingshaense had a certain degree of influence on the diversity,richness and structure of the intestinal microbial community of wireworm;the combined effects of food intake and M.pingshaense stress on the intestinal microbial community were higher than those of a single exogenous factor.
