紫檀芪对脓毒症大鼠炎性反应与心脏动作电位的抑制与调节作用 被引量:2

Inhibitory effect of pterostilbene on sepsis by repressing inflammation and regulating the cardiac action potential in rat models

摘要 目的:研究紫檀芪(PTS)对脓毒症大鼠炎性反应和心脏动作电位的作用。方法:通过盲肠结扎穿刺术建立大鼠脓毒症模型。将大鼠随机分为三组(每组10只):假手术组、脓毒症组和PTS组(腹腔注射25 mg/kg),24 h后处死大鼠,采集心肌进行心脏动作电位检测和组织病理学检查;ELISA法测定血清中肿瘤坏死因子(TNF)-α、白细胞介素(IL)-6、肌酸激酶同工酶(CK-MB)及IgE含量。结果:与假手术组比较,脓毒症组中TNF-α、IL-6、CK-MB、IgE含量均明显升高,差异有统计学意义(P<0.05),脓毒症组心房、心室动作电位时间明显长于假手术组,差异有统计学意义(P<0.05)。PTS对脓毒症组的TNF-α、IL-6、CK-MB、IgE表达水平具有显著的抑制作用,对心房、心室动作电位具有抑制作用。结论:PTS可通过抑制炎性反应和调节心脏动作电位抑制大鼠脓毒症。 Objective To explore the effect of pterostilbene(PTS)on sepsis-induced inflammation and dysregulation of cardiac action potential in rat models.Method Sepsis model was induced by cecal ligation and puncture(CLP)in rats.The rats were randomly divided into three groups(10 rats in each group):sham group,CLP group and PTS groups(intraperitoneal injection 25 mg/kg).The rats were killed after CLP for 24 h.Myocardium tissues were collected for pathological analysis and cardiac action potential detection.The content of TNF-α,IL-6,CK-MB and IgE in serum were measured by ELISA assay.Results Compared with sham group,the content of TNF-α,IL-6,CK-MB and IgE in CLP group were all promoted markedly(P<0.05).The time of atrial ventricular action potential in the CLP group was longer than that of the sham group(P<0.05).However,the administration of PTS exhibited significant inhibitory effect on the levels of TNF-α,IL-6,CK-MB and IgE,and inhibitory effects on atrial ventricular action potential in the CLP group.Conclusion The regulatory effect of PTS on inflammation and cardiac action potential leading to sepsis inhibition in rat models.
