摘要
目的:运用网络药理学方法,研究余甘子的有效成分和作用机制。方法:从中药系统药理学数据库和分析平台检索余甘子的生物活性成分、作用靶点和相关疾病,利用STRING数据库研究靶点间的相互作用关系,利用DAVID数据库分析GO和KEGG,利用Cytoscape及Hiplot软件建立“化合物-靶点”网络、蛋白质相互作用网络(PPI)、“化合物-靶点-疾病-通路”网络进行基因本体(GO)和KEGG富集分析。结果:筛选得到12个有效生物活性成分、59个靶点、107种疾病和113条通路,基因功能分析得到生物学过程(BP)相关通路479条、细胞成分(CC)相关通路44条、分子功能(MF)相关通路78条。结论:余甘子通过广泛作用于多条细胞代谢通路,调节细胞增殖、分化、凋亡的过程,发挥抗炎、抗癌活性作用。网络药理学研究有助于揭示余甘子多成分作用的协同机制。
Objective To study the effective ingredients and mechanism of emblic leafflower fruit with pharmacological method.Methods The association of interactions between targets of emblic leafflower fruit by retrieving the bioactive ingredients,acting targets and related diseases from the Systematic Pharmacology Database of Traditional Chinese Medicine and its analysis platform based on the STRING Database.The gene ontology and Kyoto encyclopedia of genes and genomes were analyzed on the basis of DAVID Database.The enriched gene ontology and Kyoto encyclopedia of genes and genomes were analyzed by establishing"compounds―targets"network,protein interaction pathway,and"compounds―targets―diseases―pathways"network based on Cytoscape and Hiplot respectively.Results Twelve effective bioactive ingredients,59 targets,107-related diseases and 113 pathways of emblic leafflower fruit were detected.Gene function analysis showed 497 biological process-related pathways,44 cellular component-related pathways and 78 molecular function-related pathways of emblic leafflower fruit.Conclusion Emblic leafflower fruit brings its anti-inflammation and anticancer activity effects into play by widely acting on cellular metabolism pathways and regulating cell proliferation,differentiation and apoptosis processes.Studies on network pharmacology help to reveal the synergistic mechanism of ingredients in emblic leafflower fruit.
