山羊DPYD和NAV3基因多态性与产羔性能的关联分析

摘要 为分析二氢嘧啶脱氢酶(DPYD)和神经导航因子3(NAV3)基因的单核苷酸多态性与山羊产羔性能的关联性,通过MassARRAY■SNP分型技术对DPYD基因和NAV3基因的4个候选位点进行分型,探究其在云上黑山羊、济宁青山羊、辽宁绒山羊3个群体中的遗传学特征,并统计其与云上黑山羊的产羔性能(产羔数、初生窝重、断奶窝重)的关联程度。群体遗传学分析表明,DPYD基因g.74670682 C>T位点和NAV3基因g.7247898 A>G位点在云上黑山羊、济宁青山羊、辽宁绒山羊中为低度多态(PIC<0.25);NAV3基因g.7078872 G>C位点在云上黑山羊和济宁青山羊中为低度多态,而在辽宁绒山羊中为中度多态(0.25<PIC<0.5);NAV3基因g.7350407 T>C位点在云上黑山羊和济宁青山羊中为中度多态,而在辽宁绒山羊中为低度多态。卡方检验表明,NAV3基因g.7247898 A>G位点在济宁青山羊、辽宁绒山羊中处于Hardy-Weinberg不平衡状态(P<0.05);NAV3基因g.7350407 T>C位点在云上黑山羊、济宁青山羊中处于Hardy-Weinberg不平衡状态(P<0.05),其余位点均处于Hardy-Weinberg平衡状态(P>0.05)。云上黑山羊的关联分析结果显示,DPYD基因g.74670682 C>T位点对产羔数无显著影响(P>0.05);NAV3基因g.7078872 G>C位点和g.7247898 A>G位点对产羔数、窝重均无显著影响(P>0.05);NAV3基因g.7350407 T>C位点对产羔数无显著影响(P>0.05),对窝重有显著影响(P<0.05);NAV3基因g.7350407 T>C位点中CC基因型个体的初生窝重显著高于CT基因型和TT基因型个体(P<0.05),TT基因型个体的初生窝重显著高于CT基因型个体(P<0.05),CC基因型个体的断奶窝重显著高于CT基因型个体(P<0.05)。综上所述,NAV3基因g.7350407 T>C位点适合作为云上黑山羊窝重选择的分子标记。 This article aims to analyze the association between single nucleotide polymorphisms of dihydropyrimidine dehydrogenase(DPYD)and neuro navigation factor 3(NAV3)genes and kidding performance in goats.MassARRAY^@SNP typing technology was applied to genotype the 4 candidate sites of DPYD and NAV3 genes and explore their genetic characteristics in Yunshang Black goat,Jining Gray goat and Liaoning Cashmere goat.And count the degree of correlation between it and the kidding performance of Yunshang Black goats(litter size,litter weight at birth and litter weight at weaning).Population genetic analysis showed that the DPYD gene g.74670682 C>T and NAV3 gene g.7247898 A>G locus was low polymorphism in Yunshang Black goats,Jining Gray goats and Liaoning Cashmere goats(PIC<0.25);the NAV3 gene g.7078872 G>C locus was low polymorphism in Yunshang Black goats and Jining Gray goats,while it was moderately polymorphic in Liaoning Cashmere goats(0.25<PIC<0.5);the NAV3 gene g.7350407 T>C locus was moderately polymorphic in Yunshang Black goats and Jining Gray goats,and It was low polymorphism in Liaoning Cashmere goats.The chi-square test showed that NAV3 gene g.7247898 A>G locus was under Hardy-Weinberg disequilibrium in Jining Gray goat and Liaoning Cashmere goats(P<0.05);The NAV3 gene g.7350407 T>C locus was under Hardy-Weinberg disequilibrium in Yunshang Black goat and Jining Gray goat(P<0.05),but the remaining sites were in Hardy-Weinberg equilibrium(P>0.05).The results of association analysis of Yunshang Black goats showed that there were no significant correlation between the DPYD gene g.74670682 C>T locus and the litter size;there was no significant correlation between NAV3 gene g.7078872 G>C,g.7247898 A>G site and litter size and litter weight(P>0.05);there was no significant correlation between NAV3 gene g.7350407 T>C site and litter size,but had a significant correlation with litter weight(P<0.05).The litter weight at birth of CC was higher than CT and TT(P<0.05),the litter weight at birth of TT was higher than CT(P<0.05)and the litter weight at weaning of CC was higher than CT(P<0.05)in NAV3 gene g.7350407 T>C site.In summary,the g.7350407 T>C locus at NAV3 gene can be suitable as a molecular marker for litter weight in Yunshang Black goats.
