山羊MED27和SCTR基因多态性与产羔性能的关联分析

摘要 旨在分析中介因子复合体27(Mediator complex 27,MED27)和分泌素受体基因(SCTR)的多态性与云上黑山羊产羔性能的关联性。利用Sequenom MassARRAY■SNP技术对MED27和SCTR基因的4个候选位点进行分型,探索其在云上黑山羊(n=544)、济宁青山羊(n=133)和辽宁绒山羊(n=91)3个群体中的遗传学特征,并分析其与云上黑山羊产羔性能(包括产羔数、初生窝重和断奶窝重)的关联性。结果表明,4个候选位点在不同繁殖力品种中的基因型分布无显著差异(P>0.05)。在云上黑山羊中,MED27基因的g.101629748G>A位点AG型个体的产羔数和断奶窝重显著高于GG型个体(P<0.05),该候选位点对初生窝重无显著影响(P>0.05);SCTR基因g.64833692C>A位点CC型个体的产羔数显著高于CA型个体(P<0.05),g.64877073C>T位点和g.64889937G>A位点对产羔数和断奶窝重无显著影响(P>0.05)。以上结果提示,MED27基因的g.101629748G>A位点和SCTR基因的g.64833692C>A位点可作为云上黑山羊产羔数选择的潜在分子标记,但SCTR基因的g.64833692C>A位点不适合窝重选择。 This study was performed to analyze the association between the polymorphism of Mediator complex 27(MED27)and SCTR genes and kidding performance of Yunshang Black goats(YS).A MassARRAY'system was used for genotyping of the four candidate loci of MED27 and SCTR,of which the genetic characteristics of YS(n=544),Jining Gray(JN,n=133)and Liaoning Cashmere goats(LN,n=91)were explored,and whose association with kidding performance including litter size,litter weight at birth and at weaning was also analyzed.The genotype distributions of four candidate loci in breeds with different fecundity were no significantly different(P>0.05).The association analyses of YS showed a striking elevated kidding number and litter weight at weaning of AG that GG(P<0.05)and of all loci on litter weight at birth(P>0.05),no significant effects of two loci(g.64877073C>T,g.64889937G>A)on litter size and litter weight at weaning;a significantly increased litter size of CC goats for g.64833692C>A and CA(P<0.05).These results suggested that the loci g.64833692C>A at SCTR unsuitable for litter wight were the potential markers for litter size in YS.The loci g.101629748G>A at MED27 were the potential markers for litter size and litter weight at weaning in YS.
