Role of hsa_circ_0005019 in proliferation,invasion and migration of colorectal cancer cells and its value of predicting prognosis of colorectal cancer

摘要 目的:评估环状RNA hsa_circ_0005019在结直肠癌细胞增殖、侵袭和迁移中的作用及对结直肠癌预后的影响。方法:人结肠癌细胞HCT116分为2组,分别转染hsa_circ_0005019 siRNA(siRNA组)和对照siRNA(阴性对照组)。采用CCK-8法和Transwell小室分别检测2组细胞增殖、侵袭和迁移能力。采用qRT-PCR检测结直肠癌和配对癌旁正常肠黏膜组织(n=66)中hsa_circ_0005019表达水平;根据hsa_circ_0005019表达水平的均值将结直肠癌患者分为高表达组和低表达组,绘制2组的生存曲线并比较预后,采用Cox回归分析hsa_circ_0005019对结直肠癌预后的影响。结果:与阴性对照组比较,siRNA组细胞增殖、侵袭和迁移能力增强(P<0.05)。结直肠癌组织中hsa_circ_0005019的相对表达量为(0.759±0.063),低于癌旁正常组织(1.000±0.082)(P<0.001)。生存分析显示,与低表达组相比,高表达组预后更好(P<0.05)。Cox回归分析表明在调整性别、年龄、病理学分级、TNM分期、肿瘤类型、浸润深度后,hsa_circ_0005019表达水平可以预测结直肠癌预后(HR=0.104,95%CI:0.013~0.854,P=0.035)。结论:Hsa_circ_0005019在结直肠癌中扮演抑癌基因的角色,有望成为结直肠癌预后标志物和生物治疗的新靶点。 Aim:To evaluate the role of circular RNA hsa_circ_0005019 in colorectal cancer(CRC)cell proliferation,invasion and migration and its value of predicting prognosis of CRC.Methods:Human CRC HCT116 cells were allocated into 2 groups and transfected with hsa_circ_0005019 siRNA(siRNA group)and control siRNA(negative control group),respectively.Cell proliferation,invasion and migration were detected by CCK-8 assay and Transwell chamber.qRT-PCR was used to compare the expression level of hsa_circ_0005019 in 66 CRC tissue samples and matched normal intestinal mucosa tissue samples.The patients with CRC were allocated into high expression group and low expression group according to the mean expression level of hsa_circ_0005019,and survival analysis was used to compare the prognosis of 2 groups.Cox regression analysis was used to analyze the value of hsa_circ_0005019 for predicting the prognosis of CRC.Results:Compared with the negative control group,cell proliferation,invasion and migration were increased in the siRNA group(P<0.05).The relative expression level of hsa_circ_0005019 in CRC tissue was(0.759±0.063),which was lower than that in normal tissue(1.000±0.082)(P<0.001).The results of survival analysis showed that prognosis was better in the high expression group than that in the low expression group(P<0.05).Cox regression analysis indicated that after adjusting gender,age,histological grade,TNM stage,types of tumor,and infiltration depth,the expression level of hsa_circ_0005019 was a factor for predicting the prognosis of CRC(HR=0.104,95%CI:0.013-0.854,P=0.035).Conclusion:Hsa_circ_0005019 may play a role in CRC as an anti-oncogene and is expected to be a prognostic marker and a new target for biological therapy in CRC.
