酸枣仁冻干粉对大鼠睡眠觉醒状态及能量代谢率的干预作用研究 被引量:1

摘要 目的探讨24 h持续黑暗条件下,酸枣仁冻干粉对大鼠睡眠觉醒状态及能量代谢率的影响及其机制。方法利用皮层脑电描记方法和实验动物监测系统(CLAMS),以觉醒及睡眠时间、活动次数、摄食量、产热量为指标,探究其对能量代谢率的改善。利用酶联免疫法测定参与下丘脑内能量调控的LEP、POMC、NPY的含量变化,研究其对能量变化的机制。结果与DD组比较,MAT组白天觉醒时间减少、睡眠增多、活动次数减少、摄食量差异无显著性、产热量增多。夜晚觉醒及睡眠时间差异无显著性、活动次数差异无显著性、摄食量减少、产热量增多。大鼠体质量增加量减少。与DD组比较,MAT组下丘脑内LEP含量升高,POMC含量升高,NPY含量降低。结论酸枣仁冻干粉通过升高下丘脑内LEP、POMC含量并降低NPY的含量,从而纠正因24 h持续黑暗条件导致的大鼠睡眠觉醒状态紊乱及能量代谢率的异常。 Objective To explore the effect of Zizyphus jujubekernelfreeze-dried powder on sleep and wakefulness and energy metabolism rate in rats under 24 h continuous darkness and its mechanism.Method Using the cortical electroencephalography method and the experimental animal monitoring system(CLAMS)and taking the time of wakefulness and sleep as well as the number of activities,food intake,and caloric production as indicators to explore the function of it on improving energy metabolism rate.Theenzyme-linked immunoassay was used to determine the changes in the content of LEP,POMC,and NPY involved in energy regulation in the hypothalamus,so as to study the energy change mechanism of it.Result Compared with the DD group,the MAT group had reduced daytime awakening time,increased sleep time,decreased activities,no significant difference in food intake,and increased caloric production.There was no significant difference in night awakening and sleep time,no significant difference in the number of activities,reduced food intake,and increased caloric production.The weight gain of rats decreased;Compared with the DD group,the LEP content in the hypothalamus of the MAT group increased,the POMC content increased and the NPY content decreased.Conclusion By increasing LEP,POMC content and decreasing NPY content in hypothalamus,Zizyphus jujubekernelfreeze-dried powder corrected the disturbance of sleep and wakefulness state and abnormal energy metabolism rate of rats caused by 24 h continuous dark condition.
