摘要
目的探讨肺腺癌转移相关转录本1(MALAT1)对耐吉西他滨胰腺癌细胞SW1990(SW1990/Gem)化疗敏感性的影响。方法应用荧光定量PCR检测MALAT1在胰腺癌细胞SW1990和SW1990/Gem中的表达水平,通过小干扰RNA下调SW1990/Gem的MALAT1水平,应用CCK-8检测吉西他滨对SW1990和SW1990/Gem的半数抑制浓度(IC_(50)),应用蛋白质印迹实验检测SW1990和SW1990/Gem的自噬相关蛋白的表达水平。结果MALAT1在SW1990/Gem中的表达水平高于SW1990(P<0.01),SW1990/Gem中的Beclin1表达水平高于SW1990(P<0.01),下调MALAT1的表达后,SW1990/Gem中的Beclin1表达量下降,吉西他滨对SW1990/Gem的IC_(50)下降。结论下调MALAT1通过抑制细胞自噬增加胰腺癌细胞对吉西他滨的敏感性。
Objective To investigate the effect of metastasis-associated lung adenocarcinoma transcript 1(MALAT1)on chemotherapy sensitivity of gemcitabine-resistant pancreatic cancer cell SW1990(SW1990/Gem).Methods The expression level of MALAT1 in SW1990 and SW1990/Gem was detected by real-time PCR.Small interfering RNA was used to down-regulate the level of MALAT1 in SW1990/Gem.The 50%inhibitory concentration(IC_(50))of gemcitabine was evaluated by CCK-8 assay.The expression of autophagyrelated protein in SW1990 and SW1990/Gem were analyzed by Western blotting.Results The expression level of MALAT1 in SW1990/Gem was higher than that in SW1990(P<0.01).The protein level of Bcelin1 in SW1990/Gem was higher than that in SW1990(P<0.01).Down-regulation of MALAT1 reduced the protein level of Beclin1,and decreased the IC50 of gemcitabine in SW1990/Gem.Conclusions Down-regulation of MALAT1 increases the chemotherapy sensitivity of gemcitabine in pancreatic cancer cell via inhibiting autophagy.
