广东省巴戟天DNA条形码及遗传多样性分析 被引量:2

摘要 目的构建广东省巴戟天DNA条形码数据库以及研究广东省不同产地巴戟天的遗传多样性。方法采用DNA条形码技术对149个巴戟天样本进行分析,构建DNA条形码数据库。利用ISSR分子标记技术对广东省8个居群的巴戟天进行遗传多样性分析。结果巴戟天五个通用条形码的PCR测序成功率为psbA-trnH>rbcL>matK>ITS2>ITS,149个巴戟天样本建立了含有569条序列的数据库。研究筛选出6条ISSR引物,从巴戟天中扩增得80条条带,多态性比率为92.50%,遗传相似系数在0.5625-0.8910,表明广东省巴戟天的遗传多样性较高。ISSR标记揭示的PCoA结果、UPGMA聚类分析结果一致,八个居群分为两大支。结论广东省巴戟天具有较高的遗传多样性。 Objective To analyze Morinda officinalis’genetic diversity by constructing the DNA barcode database of the genuine medicinal materials Morinda officinalis in Guangdong Province.Methods A total of 149 samples of M.officinalis were analyzed by DNA barcoding to construct DNA barcode database.The genetic diversity of M.officinalis sampled from 8 populations in Guangdong Province was detected based on ISSR molecular marker technology.Results The success rates of PCR amplification and sequencing of five barcodes of Morinda officinalis was psbA-trnH>rbcL>matK>ITS2>ITS.149 samples of M.officinalis were used for constructing a reference DNA barcode database containing 569 sequences.After amplification with 6 pairs of ISSR primers,80 fragments were detected,of which accounting for 92.50%of the total.The Nei’s gene diversity indices(H)ranged from 0.5625 to 0.8910.This study showed that the genetic differences among Morinda officinalis were large.The results of PCoA revealed by ISSR markers were consistent with those of UPGMA cluster analysis.8 populations were divided into two groups.Conclusion These results indicate that there is a high genetic diversity of M.officinalis in Guangdong province.
