摘要
目的建立RP-HPLC-UV色谱法测定清心莲子饮的基准物质中京尼平苷酸的含量。方法采用依利特C_(18)柱(Sinochrom ODS-BP,4.6mm×200mm,5μm),0.5%的HAc溶液和甲醇进行梯度洗脱,流速控制为1.0mL·min^(-1),在254nm,柱温25℃下进样,进样量为10μL。结果京尼平苷酸的含量与色谱峰面积呈现出良好的线性关系,其线性方程为Y=11.185X+0.065(R^(2)=0.9999)。精密度、重复性和加样回收率等试验RSD均小于3%,平均加样回收率为101.05%,RSD为2.45%。结论本文建立含量测定的方法可行,操作简易,灵敏度高,适于清心莲子饮的物质基准中京尼平苷酸的含量测定,可作为清心莲子饮质量评价的方法。
OBJECTIVE A RP-HPLC-UV chromatography method was established to determine the content of geniposide in the reference substance of Qingxin Lianzi.METHODS Elite C_(18) column(Sinochrom ODS-BP,4.6mm×200mm,5μm),0.5%acetic acid solution and methanol was used for gradient elution,the flow rate was controlled to 1.0mL·min^(-1),and the column temperature was 254nm at 25℃.The injection volume is 10μL.RESULTS The content of geniposide has a good linear relationship with the chromatographic peakarea,and the linear equation is Y=11.185X+0.065(R^(2)=0.9999).The RSD of precision,repeatability and sample recovery rate were all less than 3%.The average sample recovery rate was 101.05%,and the RSD was 2.45%.CONCLUSION The method established in this paper is highly feasible,easy to operate,and highly sensitive.It is suitable for the determination of geniposide in the substance standard of Qingxin Lianzi and can be used as a method for quality evaluation of Qingxin Lianzi.
