摘要
目的:建立同时测定女贞子药材中酪醇、红景天苷、特女贞苷、松果菊苷、毛蕊花糖苷含量的高效液相色谱方法,并考察女贞子不同产地及采收期苯乙醇苷类成分的含量。方法:样品经50%甲醇回流提取,采用ZORBAX SB-C_(18)色谱柱(250 mm×4.6 mm,5μm),柱温40℃,以乙腈-0.1%磷酸溶液为流动相,梯度洗脱,流速1.0 ml·min^(-1),检测波长为224 nm(特女贞苷)、275 nm(酪醇、红景天苷)、332 nm(松果菊苷、毛蕊花糖苷)。运用聚类分析和主成分分析对数据进行统计。结果:聚类分析显示21批不同产地女贞子可分为3类,主成分分析提取出2个主成分,主成分综合评分显示湖南省郴州市安仁县、湖南省岳阳市、浙江省丽水市缙云大源镇女贞子品质较好。不同采收期含量测定结果表明5种成分总含量在12月份达到最高。结论:不同产地女贞子不同采收期女贞子5种成分含量差异较大,12月份可作为湖南省长沙市女贞子的适宜采收期,所建立的高效液相色谱色谱方法简便、准确,可用于女贞子的质量控制。
Objective:To establish an HPLC method for the simultaneous determination of tyrosol,salidroside,specnuezhenide,echinacoside and acteoside in Fructus Ligustrum Lucidi,and to investigate the best producing area and harvest time of Fructus Ligustrum Lucidi.Methods:The samples were extracted by 50%ethanol under reflux condition,and the determination was performed by using a ZORBAXSB-C_(18)column(250 mm×4.6 mm,5μm)with the column temperature at 40℃.The mobile phase was acetonitrile-0.1%phosphoric acid solution with gradient elution at the flow rate of 1.0 ml min^(-1),and the detection wavelengths were 224 nm(specnuezhenide),275 nm(tyrosol,salidroside)and 332 nm(echinacoside,acteoside).Cluster analysis and principal component analysis were used to analyze the data.Results:Cluster analysis showed that 21 batches of Ligustrum lucidum from different habitats could be divided into 3 categories,two principal components were extracted by principal component analysis.The comprehensive scores of principal components showed that the quality of Fructus Ligustrum Lucidi was better in Anren county of Chenzhou city in Hunan province,Yueyang city in Hunan province and Dayuan town in Jinyun of Lishui city in Zhejiang province.The results of content determination in different harvest periods showed that the total content of five components reached the highest in December.Conclusion:The contents of five components in Fructus Ligustrum Lucidi from different producing areas and in different harvest periods are quite different,and the month of December can be selected as the suitable harvest period of Ligustrum lucidum in Changsha of Hunan province.The established HPLC method is simple and accurate,and can be used for the quality control of Fructus Ligustrum Lucidi.
