不同配比赤芍-川芎药对配伍中11个成分提取率考察 被引量:5

Study on the Effects of Different Compatibility Ratios on the Dissolution of 11 Components in Drug Pair Paeoniae Radix Rubra and Ligusticum Chuanxiong

摘要 目的:建立赤芍-川芎药对中没食子酸、绿原酸、氧化芍药苷、芍药内酯苷、芍药苷、阿魏酸、1,2,3,4,6-O-五没食子酰葡萄糖、洋川芎内酯Ⅰ、阿魏酸松柏酯、洋川芎内酯A及藁本内酯11个成分的含量测定方法,考察不同配伍比例及配伍方式对赤芍-川芎药对中11个主要活性成分溶出量的影响。方法:采用Welch Ultimate;XBC;色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈(A)-0.1%磷酸溶液(B),梯度洗脱;检测波长为230 nm,流速为1.0 ml·min^(-1),柱温为35℃,对赤芍中5个成分(没食子酸、氧化芍药苷、芍药内酯苷、芍药苷及1,2,3,4,6-O-五没食子酰葡萄糖)进行检测;采用Boston C;色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈(A)-0.5%醋酸溶液(B),梯度洗脱;检测波长为285 nm,流速为1.0 ml·min^(-1),柱温为30℃,对川芎中6个成分(绿原酸、阿魏酸、洋川芎内酯Ⅰ、阿魏酸松柏酯、洋川芎内酯A及藁本内酯)进行检测。进一步测定赤芍-川芎药对的单煎液、单煎合并液(1∶1)以及不同配比的合煎液(3∶1,2∶1,1∶1,1∶2,1∶3)中上述11个主要活性成分的含量。结果:合煎液中各成分提取率明显大于单煎液和单煎合并液中提取率,且在赤芍-川芎药对配比为1∶1时多种成分含量最高。结论:赤芍-川芎(1∶1)的配伍比例以及合煎的配伍方式有利于11个成分在水煎液中的溶出。 Objective:To establish a method for the simultaneous determination of eleven components(gallic acid,chlorogenic acid,oxypaeoniflorin,albiflorin,paeoniflorin,ferulic acid,1,2,3,4,6-penta-O-galloy-β-D-glucose,senkyunolide I,coniferly ferulate,senkyunolide A and ligustilide)in drug pair Paeoniae Radix Rubra and Ligusticum chuanxiong,and to study the influence of compatibility proportions and compatibility methods on the dissolution of the 11 components.Methods:A Welch Ultimate;XB C;column(250 mm×4.6 mm,5μm)with a column temperature at 35℃was used,the mobile phase consisted of acetonitrile(A)-0.1%formic acid(B)(gradient elution)at a flow rate of 1.0 ml·min^(-1),and the detection wavelength was set at 230 nm for the determination of gallic acid,oxypaeoniflorin,albiflorin,paeoniflorin and 1,2,3,4,6-penta-O-galloy-β-D-glucose in Paeoniae Radix Rubra.And a Boston C;(250 mm×4.6 mm,5μm)with a column temperature at 30℃was used,the mobile phase consisted of acetonitrile(A)-0.5%acetic acid solution(B)(gradient elution)at a flow rate of 1.0 ml·min^(-1),and the detection wavelength was set at 285 nm for the determination of ferulic acid,senkyunolide I,coniferly ferulate,senkyunolide A and ligustilide in Ligusticum chuanxiong.And the eleven components were determined at different compatibility proportions,including single decoction,mixed single decoction(1∶1)and mixed decoction(1∶1,1∶2,1∶3,2∶1 and 3∶1).Results:Compared with the single decoction and mixed single decoction,when the proportion of Paeoniae Radix Rubra-Ligusticum chuanxiong was 1∶1,the contents of 11 components increased obviously in mixed decoction,and the contents of the most of components were the highest in 1∶1 drug pair.Conclusion:The mixed decoction of Paeoniae Radix Rubra-Ligusticum chuanxiong at the proportion of 1∶1 has an obvious promoting effect on the dissolution of the 11 components.
