热毒宁注射液对肺炎克雷伯菌所致幼龄小鼠肺功能损伤和免疫紊乱的调节作用 被引量:1

Effects of Reduning Injection on Lung Function Injury and Immune Disorder Induced by Klebsiella Pneumoniae in Young Mice

摘要 目的探讨热毒宁注射液对肺炎克雷伯菌诱导的幼龄小鼠肺功能损伤和免疫紊乱调节的影响。方法将小鼠随机分为5组:对照组、模型组、热毒宁注射液低中高剂量组。HE染色检测肺组织病理情况。肺组织病理学评分和W/D值评估肺损伤。RT-PCR检测Ki67、Survivin mRNA表达。Westernt印迹检测Caspase-3、Caspase-9、TGF-β和α-SMA蛋白表达。Masson染色检测肺组织纤维化。ELISA检测肺组织和外周血中iNOS、IL-10含量。结果与模型组比较,热毒宁注射液中高剂量组的肺损伤和肺纤维化明显改善,肺组织病理学评分和W/D值降低(P<0.05),Ki67和Survivin mRNA表达升高(P<0.05),Caspase-3、Caspase-9、TGF-β和α-SMA蛋白表达降低(P<0.05),组织和外周血中的iNOS含量降低(P<0.05),IL-10含量升高(P<0.05)。结论热毒宁注射液可改善肺炎克雷伯菌所致幼龄小鼠的肺功能损伤和免疫紊乱。 Objective To investigate the effects of Reduning injection on lung function injury and immune disorder induced byKlebsiella pneumoniaein young mice.Methods The mice were ran-domly divided into 5 groups:control group,model group and low-,medium-and high-dose Redun-ing injection groups.HE staining was used to detect the pathology of lung tissues.Lung histopatholog-ical score and W/D ratio were used to assess lung injury.The expression of Ki67 and Survivin mRNA was detected by RT-PCR.The expression of Caspase-3,Caspase-9,TGF-βandα-SMA proteins were detected by Western blot.Masson staining was used to determine pulmonary fibrosis.The con-tents of iNOS and IL-10 in lung tissue and peripheral blood were determined by ELISA.Results In Reduning injection groups relative to the KPN group,the lung injury and pulmonary fibrosis were significantly improved;the lung tissue score and W/D ratio were markedly decreased(P<0.05).The expression levels of Ki67 and Survivin mRNA were significantly increased(P<0.05).The ex-pression levels of Caspase-3,Caspase-9,TGF-βandα-SMA protein were profoundly decreased(P<0.05).The iNOS content in tissues and peripheral blood was significantly decreased(P<0.05).The IL-10 content was obviously increased(P<0.05).Conclusion Reduning injection can alleviate the lung function injury and immune disorder caused byKlebsiella pneumoniaein young mice.
