摘要
目的分析皮肤鳞状细胞癌(cSCC)组织中微小RNA(miRNA)-383-5p、驱动蛋白家族成员3B(KIF3B)mRNA的表达及临床意义。方法选取2016年1月—2021年8月华中科技大学同济医学院附属同济医院皮肤科收治手术切除cSCC患者49例,qRT-PCR检测癌组织和癌旁组织中miR-383-5p、KIF3B mRNA表达。Pearson相关性分析cSCC癌组织中miR-383-5p与KIF3B mRNA表达的相关性。分析miR-383-5p、KIF3B mRNA表达与cSCC患者临床病理参数的关系。ROC曲线分析miR-383-5p、KIF3B mRNA表达对cSCC发生的预测价值。结果与癌旁组织比较,癌组织中miR-383-5p表达降低,KIF3B mRNA表达升高(t=31.868、33.517,P均<0.001)。Pearson相关性分析显示,cSCC癌组织中miR-383-5p与KIF3B mRNA表达呈负相关(r=-0.573,P<0.001)。miR-383-5p、KIF3B mRNA表达与cSCC患者TNM分期、侵袭程度、淋巴结转移有关(P<0.01)。ROC曲线显示,miR-383-5p、KIF3B mRNA及二者联合预测cSCC发生的曲线下面积(AUC)分别为0.807、0.800、0.905,敏感度分别为0.837、0.878、0.735,特异度分别为0.653、0.612、0.918。二者联合预测cSCC发生的AUC大于单独预测(Z=2.936、3.220,P=0.003、0.001)。结论cSCC癌组织中miR-383-5p低表达,KIF3B mRNA高表达,与TNM分期、侵袭程度、淋巴结转移有关,二者可能共同参与cSCC发生发展,可作为cSCC发生预测指标。
Objective To analyze the expression and clinical significance of microRNA(miRNA)-383-5p and kinesin family member 3B(KIF3B)mRNA in cutaneous squamous cell carcinoma(cSCC)tissues.Methods From January 2016 to August 2021,49 patients with cSCC who underwent surgical resection in the Department of Dermatology,Tongji Hospital,Tongji Medical College,Huazhong University of Science and Technology were selected.The qRT-PCR was used to detect the mRNA expression of miR-383-5p and KIF3B in cancer tissue and adjacent tissue.Pearson correlation analysis of the correlation between miR-383-5p and KIF3B mRNA expression in cSCC cancer tissues.The relationship between miR-383-5p and KIF3B mRNA expression and clinicopathological parameters in cSCC patients was analyzed.ROC curve analysis of the predictive value of miR-383-5p and KIF3B mRNA expression on the occurrence of cSCC.Results Compared with the adjacent tissue,the expression of miR-383-5p in the cancer tissue was decreased,and the expression of KIF3B mRNA was increased(t=31.868,33.517,both P<0.001).Pearson correlation analysis showed that miR-383-5p was negatively correlated with KIF3B mRNA expression in cSCC cancer tissues(r=-0.573,P<0.001).The expression of miR-383-5p and KIF3B mRNA was related to TNM stage,degree of invasion and lymph node metastasis in cSCC patients(P<0.01).The ROC curve showed that the area under the curve(AUC)of miR-383-5p,KIF3B mRNA and their combination in predicting the occurrence of cSCC were 0.807,0.800,0.905,the sensitivity was 0.837,0.878,0.735,and the specificity was 0.653,0.612,0.918.The AUC of the two combined to predict the occurrence of cSCC was greater than the independent prediction(Z=2.936,3.220,P=0.003,0.001).Conclusion The low expression of miR-383-5p and the high expression of KIF3B mRNA in cSCC cancer tissues are related to TNM stage,degree of invasion,and lymph node metastasis.
