黄芪超微粉对环磷酰胺致免疫失衡小鼠免疫功能的影响 被引量:8

摘要 目的:观察黄芪超微粉对环磷酰胺免疫失衡小鼠免疫功能的影响。方法:将5~6周龄昆明种小鼠60只随机分为对照组、模型组、黄芪饮片组(HQY,10g/kg)、黄芪超微粉高剂量组(HQFH,10g/kg)、黄芪超微粉中剂量组(HQFM,2g/kg)、黄芪超微粉低剂量组(HQFL,0.4g/kg),每组10只,雌雄各半。模型组和各药物治疗组小鼠采用80mg/kg环磷酰胺腹腔注射建立小鼠免疫失衡模型,连续注射4天,然后每周强化1次,共强化2次,各药物治疗组小鼠于环磷酰胺腹腔注射当日开始灌胃给药,所有小鼠于末次强化后第1天取材,进行指标检测。采用器官重量法测胸腺指数和脾指数;采用MTT法测脾淋巴细胞增殖能力;采用Elisa法测量脾细胞上清IL-2和INF-γ细胞因子含量;采用流式细胞仪检测外周血和脾脏免疫细胞分群。结果:与对照组比较,模型组小鼠的胸腺指数明显降低(P<0.05),脾指数增高(P<0.05),脾细胞刺激指数降低(P<0.05),脾细胞细胞因子含量降低(P<0.05),外周血和脾脏CD3^(+)细胞增多(P<0.05),CD19^(+)、CD314^(+)细胞减少(P<0.05);与环磷酰胺组比较,黄芪饮片及黄芪超微粉各剂量组小鼠的胸腺指数明显升高(P<0.05),脾指数降低(P<0.05),脾细胞刺激指数升高(P<0.05),脾细胞细胞因子含量增高(P<0.05),外周血和脾脏CD3^(+)细胞减少(P<0.05),CD19^(+)、CD314^(+)细胞升高(P<0.05)。结论:黄芪超微粉可以对抗环磷酰胺所致小鼠的免疫失衡,调节小鼠免疫功能。 Objective:To observe the effect of Astragalus superfine powder on immune function of mice with cyclophosphamide immune imbalance.Methods:60 KM mice aged 5 to 6 weeks were randomly divided into control group,model group,Astragalus Decoction group(HQY,10 g/kg),high dose group(HQFH,10 g/kg),medium dose group(HQFM,2 g/kg),low dose group(HQFL,0.4 g/kg)of Astragalus superfine powder,10 mice of half male and half female in each group,The immune imbalance model of mice was established by intraperitoneal injection of 80 mg/kg cyclophosphamide for 4 consecutive days in mice of model and treatment groups,and then strengthened once a week for a total of 2 times.The mice in each treatment group were administered by gavage on the day injected intraperitoneally with cyclophosphamide.All mice were killed on the first day after the last cyclophosphamide strengthening,and the samples were taken for detection.Thymus index and spleen index were measured by method of organ weight;The proliferation of splenic lymphocytes was measured by MTT assay;The content of IL-2 and INF-γin Spleen cell supernatant were measured by ELISA method.Flow cytometry was used to detect the distribution of immune cells in peripheral blood and spleen.Results:Compared with the control group,the thymus index was significantly decreased(P<0.05),the spleen index was increased(P<0.05),the spleenocyte stimulating index was decreased(P<0.05),and the cytokine content of spleenocytes was decreased(P<0.05),CD3^(+)cells in peripheral blood and spleen increased(P<0.05),while CD19^(+)and CD314^(+)cells decreased in model group(P<0.05);Compared with the model group,the thymus index of mice increased significantly(P<0.05),while the spleen index decreased(P<0.05),the stimulation index of spleenocytes increased(P<0.05),the cytokine content in the supernatant of spleenocytes increased(P<0.05),CD3^(+)cells in peripheral blood and spleen decreased(P<0.05),CD19^(+)and CD314^(+)cells increased(P<0.05)in the Astragalus decoction pieces and Astragalus superfine powder groups.Conclusion:Astragalus superfine powder can antagonize the immune imbalance induced by cyclophosphamide and regulate the immune function of mice.
