卵巢癌组织中miR-183表达及对癌细胞增殖和凋亡影响 被引量:1

Expression of microRNA-183 in ovarian tissue and its effect on the proliferation and apoptosis of ovarian cancer cells

摘要 目的:探讨微小RNA-183(miR-183)在人卵巢癌细胞中的表达及其在卵巢癌细胞增殖和凋亡中的作用。方法:采用荧光实时定量PCR检测癌组织和癌旁组织中miR-183基因表达。噻唑蓝法检测miR-183模拟物导入人卵巢腺癌细胞(SKOV3)和人卵巢癌细胞(OVCAR3)细胞系的癌细胞增殖情况。采用流式细胞仪检测细胞凋亡情况。结果:卵巢癌组织细胞中MIR-183(2.75±1.02)表达低于癌旁组织(5.43±1.27)(P<0.05)。MTT和流式细胞仪结果显示,与对照组相比,转染组卵巢癌细胞增殖活性降低,SKOV3和OVCAR3凋亡率[(18.13±1.85)%,(21.11±2.16)%]升高(P<0.05)。结论:miR-183与卵巢癌的发生有关,miR-183的高水平表达能够抑制卵巢癌细胞增殖,促进卵巢癌细胞凋亡。 Objective:To investigate the expression of microRNA-183 in ovarian cancer cells,and to study its effect on the proliferation and apoptosis of ovarian cancer cells.Methods:Fluorescence quantitative real-time PCR(RT-QPCR)was used to detect the expression level of Mir-183 gene in ovarian cancer and paracancer tissues.The proliferation of human ovarian cancer cells(SKOV3)and human ovarian cancer ovarian cancer cell(OVCAR3)lines introduced by Mir-183 simulant was determined by the thiazolam(MTT)assay.Cell apoptosis was detected by flow cytometry.Results:The mir-183 expression level(2.75±1.02)in ovarian cancer tissue was significant lower than that(5.43±1.27)in paracancer tissue(P<0.05).MTT and flow cytometry results showed that compared with that of cells without transfection,the proliferative activity of ovarian cancer cells after transfection had decreased significantly(P<0.05),but the apoptosis rates of SKOV3 and OVCAR3(18.13±1.85%and 21.11±2.16%)had increased significantly(P<0.05).Conclusion:The expression level of microRNA-183 is closely related to the occurrence of ovarian cancer,and the high expression level of microRNA-183 can inhibit the proliferation of ovarian cancer cells and promote the apoptosis of ovarian cancer cells.
