Optimization of Culture Medium of Serendipita indica Based on Response Surface Methodology

摘要 印度梨形孢(Serendipita indica)是一种能在人工培养基上培养的根内生真菌。为了提高S.indica的生长速率,进一步优化S.indica培养基。本研究通过单因素实验探究6种影响因子对S.indica生长的影响,并通过响应面法对S.indica培养基进行精细优化。结果表明,S.indica菌株生长的最适温度为31±2℃,最适pH为6.9±0.2,最佳碳源为蔗糖,最佳氮源为NaNO_(3),最佳无机盐为MgSO_(4)·7H_(2)O。应用RSM最终确定S.indica菌株优化后的培养基配方如下:MgSO_(4)·7H_(2)O为0.824 g/L、KH_(2)PO_(4)为1.118 g/L、NaNO_(3)为1.055 g/L、NaCl为0.03 g/L、CaCl_(2)为0.05 g/L、蔗糖为10 g/L;在此条件下,S.indica的平均生长速率为1.8083 cm/d,远大于原MMN培养基继代培养的平均生长速率。该研究通过RSM优化S.indica培养基成分,提高S.indica生长速率,为S.indica菌株规模化生产并在田间试验中的应用奠定基础。 The root endophytic fungus Serendipita indica can be cultured on artificial medium.The present study aims at optimizing the medium to enhance the growth rate of S.indica.In this experiment,the single-factor experiment was selected for evaluating the effect of six parameters on the growth rate of S.indica,and the optimization design of the medium was performed by the response surface methodology(RSM).The results showed that the optimum temperature was 31±2℃,the optimum pH was 6.9±0.2,the best carbon source was sucrose,the best nitrogen source was NaNO_(3),and the best inorganic salt was MgSO_(4)·7H_(2)O.The optimal medium formulation was determined by RSM as following:MgSO_(4)·7H_(2)O 0.824 g/L,KH_(2)PO_(4)1.118 g/L,NaNO_(3)1.055 g/L,NaCl 0.03 g/L,CaCl_(2)0.05 g/L and sucrose 10 g/L.In the optimal medium,the average growth rate of S.indica reached 1.8083 cm/d,which was greatly faster than that in the original MMN medium.In this study,medium optimization based on RSM improved the growth rate of S.indica,which laid a foundation for the large-scale production of S.indica and its application in field trials.
