日粮蛋氨酸水平对大骨鸡肌肉SLC12A5、Musclin mRNA表达影响的研究

摘要 试验旨在研究日粮蛋氨酸水平大骨鸡腿肌和胸肌SLC12A5、Musclin mRNA表达量的影响。选择240只16周龄公鸡,随机分为3组,每组10个重复,每个重复8只。H组、M组和L组日粮中蛋氨酸(Met)水平分别为0.42%、0.34%、0.25%,其中M组为对照组。试验期8 w。结果显示,H组各周龄公鸡血清Met含量均极显著低于M组(P<0.01);各周龄L组公鸡腿肌Musclin mRNA表达量均极显著高于H组(P<0.01);第16 w,H组、L组公鸡胸肌Musclin mRNA表达量极显著均低于M组(P<0.01),H组显著高于L组(P<0.05);第16 w,L组公鸡腿肌SLC12A5 mRNA表达量极显著高于H组(P<0.01),L组显著低于M组(P<0.05);第24 w,H组胸肌SLC12A5 mRNA表达量显著低于其他两组(P<0.05)。研究表明,日粮Met水平能够影响血清Met含量和胸肌和腿肌的蛋白质合成;但SLC12A5基因表达量与Met水平不呈剂量-效应关系。 The experiment was to study the effect of dietary methionine levels on expression of SLC12 A5 and Musclin mRNA in thigh and breast muscles of Dagu chickens.240 healthy 16-week-old roosters were selected and randomly divided into three groups,with ten replicates in each group and eight chickens in each replicate.The methionine(Met)level in group H,group M and group L was 0.42%,0.34%and 0.25%,respectively.Group M was the control group.The trial period was eight weeks.The results showed that the serum Met content in group H was extremely lower than that in group M(P<0.01).The mRNA expression of Musclin in thigh muscle of rooster in group L was extremely higher than that in group H(P<0.01).At the 16 th week,the mRNA expression of Musclin in breast muscle of male chicken in group H and group L was extremely lower than that in group M(P<0.01),and that in group H was significantly higher than that in group L(P<0.05).At the 16 th week,SLC12 A5 mRNA expression in group L was extremely higher than that in group H(P<0.01),and that in group L was significantly lower than that in group M(P<0.05).The SLC12 A5 mRNA expression in breast muscle of group H was significantly lower than that of the other two groups at the 24 th week(P<0.05).The experiment indicates that dietary Met level can affect serum Met content and protein synthesis of breast muscle and leg muscle,but there is no dose-effect relationship between SLC12 A5 gene expression and Met level.
