白细胞介素-1受体拮抗剂等位基因2串联重复多态性与原发性膝骨关节炎易感性的关系 被引量:2

Association of interleukin-1 receptor antagonist allele 2 tandem repeat polymorphism and susceptibility to primary knee osteoarthritis

摘要 目的分析白细胞介素-1受体拮抗剂等位基因2(IL-1RN*2)串联重复多态性与原发性膝关节骨关节炎(PKOA)易感性的关系。方法PKOA患者244例(观察组),其中变异型者65例(含有IL-1RN*2型等基因)、野生型者179例(含有IL-1RN*1型等位基因),另选233例健康查体者作对照(对照组),比较IL-1RN各个基因型、等位基因的出现频率及与PKOA高发生率的相关性。结果观察组1/1基因型176例(72.1%)、1/2基因型53例(21.7%)、2/2基因型12例(4.90%)、1/4基因型3例(1.30%),对照组分别为169例(72.5%)、50例(21.5%)、10例(4.30%)、4例(1.70%),两组比较,P均>0.05。观察组1等位基因408例(83.6%)、2等位基因77例(15.8%)、4等位基因3例(0.6%),对照组分别为392例(84.1%)、70例(15.0%)、4例(0.9%),两组比较,P均>0.05。风险性非条件Logistic回归分析发现,IL-1RN基因变异型(1/2和2/2)发生PKOA的风险是非变异基因型(1/1+1/3+1/4)的1.955倍(OR=1.955,95%CI=0.635~5.437),其中携带IL-1RN*2基因位点发生PKOA的风险是IL-1RN*1基因位点的1.589倍(OR=1.589,95%CI=0.791~3.016)。结论IL-1RN*2基因串联重复多态性与胶东地区PKOA易感性无明显相关性。 Objective To analyze the correlation between interleukin-1 receptor antagonist allele 2(IL-1 RN*2)tandem repeat polymorphism and susceptibility to primary knee osteoarthritis(PKOA).Methods We selected 244 PKOA patients(observation group),including 65 patients with variant type(including IL-1 RN*2 gene)and 179 patients with wild type(including IL-1 RN*1 allele),and 233 healthy subjects(control group),and then compared the occurrence frequencies of IL-1 RN genotypes and alleles and their correlation with the high incidence of PKOA.Results In the observation group,there were 176(72.1%)cases of 1/1 genotype,53(21.7%)cases of 1/2 genotype,12(4.90%)cases of 2/2 genotype,and 3(1.30%)cases of 1/4 genotype;there were 169 cases(72.5%),50 cases(21.5%),10 cases(4.30%)and 4 cases(1.70%)in the control group,respectively;there was no statistical significance between the two groups(all P>0.05).There were 408 cases(83.6%)of allele 1,77 cases(15.8%)of allele 2,and 3 cases(0.6%)of allele 4 in the observation group,and 392 cases(84.1%),70 cases(15.0%)and 4 cases(0.9%)in the control group,respectively;there was no significant difference between the two groups(all P>0.05).However,nonconditional Logistic regression analysis showed that the risk of PKOA in IL-1 RN variant(1/2 and 2/2)was 1.955 times that of non-variants(1/1+1/3+1/4)(OR=1.955,95%CI=0.635-5.437);the probability of PKOA with IL-1 RN*2 locus was 1.589 times that of IL-1 RN*1 locus(OR=1.589,95%CI=0.791-3.016).Conclusion There was no significant correlation between IL-1 RN*2 gene tandem repeat polymorphism and the susceptibility to PKOA in Jiaodong area.
