摘要
以六妹羊肚菌(Morchella sextelata)GZCC0287菌株为试材,采用单因素试验、Plackett-Burman试验、最陡爬坡试验、响应面法,研究了碳源、氮源、无机盐、CMC-Na对六妹羊肚菌丝生物量及菌丝球浓度的影响,以期为羊肚菌液体菌种生产、栽培种、工业化生产提供参考依据。结果表明:红糖、酵母粉、KH_(2)PO_(4)、MgCl_(2)·6H_(2)O为最佳碳源、氮源和无机盐,最佳培养基配方为红糖50.49g·L^(-1)、酵母粉10g·L^(-1)、KH_(2)PO_(4)1g·L^(-1)、MgCl_(2)·6H_(2)O 2.37g·L^(-1)、CMC-Na 10.11g·L^(-1),菌丝生物量达到(5.21±0.45)g,菌丝球浓度达到(2567±127)个·mL^(-1),以1000个·mL^(-1)浓度接种栽培种,发菌快,长势好,满袋时间10~15d。
Taking M.sextelata strain GZCC0287as the test material,carbon source,nitrogen source,inorganic salt and sodium carboxymethylcellulose(CMC-Na)were investigate by single factor design,Plackett-Burman design,and central composite design for biomass weight and concentration of mycelium pellet,respectively,in order to provide an effective way for produce the liquid spawns,solid strain and industrial fermentation.The results showed that brown sugar,yeast powder,KH_(2)PO_(4),MgCl_(2)·6H_(2)O were optimal carbon source,nitrogen source and inorganic salt individually.And the optimized medium combination was brown sugar 50.49g·L^(-1),yeast powder 10g·L^(-1),KH_(2)PO_(4)1g·L^(-1),MgCl_(2)·6H_(2)O 2.37g·L^(-1),CMC-Na 10.11g·L^(-1).Under this condition,the maximum biomass weight was(5.21±0.45)g and the concentration of mycelia pellets was 2567±127per milliliter.The mycelium of Morchella spp.filled the cultivation bag for 10days to 15days by inoculated liquid seed 5mL for 1000per milliliter.
