复脉康胶囊对缓慢型心律失常大鼠心肌细胞Na^(+)/K^(+)-ATP酶及α_(1)、α_(2)亚基表达的影响

摘要 目的研究复脉康胶囊对缓慢型心律失常大鼠心肌细胞Na^(+)/K^(+)-ATP酶(NKA酶)及α_(1)、α_(2)亚基蛋白表达的影响,探讨复脉康胶囊对缓慢型心律失常的作用机制。方法采用尾静脉处注入异搏定的方法制备缓慢型心律失常大鼠模型,将造模成功的60只SD大鼠随机均分为空白组、模型组、复脉康胶囊组(低、中、高剂量组)、心宝丸组。采用NKA酶活性测定试剂盒测定各组大鼠心肌中NKA酶的活性,Western blot法检测心肌细胞内钠泵α_(1)、α_(2)亚基蛋白的表达。结果与模型组比较,复脉康胶囊组、心宝丸组的NKA酶活性明显升高。与模型组比较,复脉康胶囊组、心宝丸组的钠泵α_(1)、α_(2)亚基蛋白的表达均明显升高。结论缓慢型心率失常大鼠心肌细胞NKA酶的活性及α_(1)、α_(2)亚基蛋白的表达明显下降,复脉康胶囊能提高其NKA酶的活性及α_(1)、α_(2)亚基蛋白的表达量,提示复脉康胶囊可能通过此机制来治疗缓慢型心律失常患者。 OBJECTIVE To study the effect of Fumaikang capsule(FC)on Na^(+)/K^(+)-ATPase(NKA)activity and the expression ofα_(1)、α_(2) subunit protein in myocardial cells of rats with chronic cardiac arrhythmias,and to explore the mechanism of FC on chronic cardiac arrhythmias.METHODS The chronic cardiac arrhythmias rat model was established by injecting verapamil into the tail vein.The 60 SD rats that were successfully modeled were randomly and averagely divided into blank group,model group,FC groups(low-dose,medium dose and high dose group),and the Xinbao pill(XBP)group.The NKA activity assay kit was used to determine the NKA activity in the myocardium of rats in each group.Western blot was used to detect the protein expression of sodium pumpα;andα;subunits in myocardial cells.RESULTS Compared with the model group,the NKA activity of the FC group and the XBP group were significantly increased.Compared with the model group,the expression of sodium pumpα;andα;subunit protein in the FC group and the XBP group were significantly increased.CONCLUSION NKA activity andα_(1)、α_(2)subunit protein expression in myocardial cells of rats with chronic cardiac arrhythmias are significantly decreased.FC can increase its NKA activity andα_(1)、α_(2)subunit protein expression,suggesting that FC may treat patients with chronic cardiac arrhythmias through those mechanisms.
